Implementing organ-on-chip in a next-generation risk assessment of chemicals: a review

Katharina S. Nitsche et al.

Summary: Organ-on-chip (OoC) technology has the potential to revolutionize the Next-Generation Risk Assessment of consumer products and chemicals by combining human cell culturing and dynamic microfluidics. The success of this technology depends on robust microfluidic devices and organ tissue models. Recent advancements in manufacturing and tissue cultivation protocols have bridged the gaps towards implementing OoC technology. Next-Generation Risk Assessment is an exposure-led and hypothesis-driven approach that utilizes human exposure information and non-animal toxicological testing. This review critically examines commercial OoC devices and recent studies on tissue culture models for Next-Generation Risk Assessment.

Safety and fate of nanomaterials in food: The role of in vitro tests

Miguel Xavier et al.

Summary: The use of nanomaterials in food and feed production is expected to increase, with the EFSA recently publishing new guidelines for their risk assessment. However, few publications cover the extensive list of in vitro studies and parameters recommended by EFSA, but some address specific sections comprehensively. Emerging technologies like organ-on-chip may accelerate toxicokinetics studies and improve in vitro-in vivo correlation.

Current EU regulatory requirements for the assessment of chemicals and cosmetic products: challenges and opportunities for introducing new approach methodologies

Francesca Pistollato et al.

Summary: EU regulations advocate for changing the way toxicity testing is conducted, emphasizing the use of non-animal methods. While some endpoints can be addressed with non-animal approaches, reliance on animal testing still exists for certain systemic effects. Challenges are faced in introducing alternative methods into regulatory testing practice.

An integrated approach to testing and assessment of high aspect ratio nanomaterials and its application for grouping based on a common mesothelioma hazard

Fiona Murphy et al.

Summary: The development of an Integrated Approach to Testing and Assessment (IATA) aimed to support the grouping of different types of High Aspect Ratio Nanomaterials (HARNs) based on their potential to cause mesothelioma. The approach is hypothesis-driven, linking critical physicochemical descriptors for NFs to defined fate/toxicokinetic and hazard outcomes.
