A chromosome-level genome assembly of the European Beech (Fagus sylvativa) reveals anomalies for organelle DNA integration, repeat content and distribution of SNPs

Summary: This study presents a chromosome-level assembly of the genome of a 300-year-old European Beech individual, revealing the characteristics and distribution of its nuclear genome. Multiple insertions of the chloroplast genome were observed, with one region on chromosome 11 showing random insertions of fragments covering the entire chloroplast genome. Telomeric repeats were found at the ends of most assembled chromosomes, while centromeric repeats were scattered throughout the genome. The distribution of SNPs, repeat elements, and duplicated genes was unevenly distributed, with a major anomaly on chromosome 4.

The pip1s Quintuple Mutants Demonstrate the Essential Roles of PIP1s in the Plant Growth and Development of Arabidopsis

Summary: The study used CRISPR/Cas9 system to mutate PIP1;4 and PIP1;5 genes in a triple mutant, generating a quintuple mutant. The pip1s(-) mutants exhibited smaller leaves and flowers, fewer developed siliques, shorter silique length, and decreased seed production compared to the wild-type plant. The mutant also showed significantly lower pollen germination rate and deformed pollen outer wall, along with transcriptomic changes in key genes related to leaf, flower, and pollen development.
