Development of human alveolar epithelial cell models to study distal lung biology and disease

Summary: In this study, we successfully established alveolar epithelial cell lines using de-identified human remnant transplant lungs. These cell lines were validated in both 2D and 3D cultures. In 3D culture, the cells formed alveolar-like organoids and expressed mature alveolar epithelial cell markers. Single-cell RNA sequencing revealed increased cellular heterogeneity and induction of cytokine and lipoprotein signaling in the 3D organoids.

Summary: Exposure to particulate matter pollution, particularly ultrafine particles, has damaging effects on the human brain, especially among young individuals. Current standards in the US do not explicitly regulate ultrafine particles, despite growing evidence of their significant health impacts. A case study in Metropolitan Mexico City has shown the presence of neurodegenerative diseases and nanoparticles in the brains of young residents. The US needs to adopt more comprehensive regulations and interventions to mitigate ultrafine particle emissions and prevent associated neurological outcomes.

Low-Dose Cadmium Potentiates Metabolic Reprogramming Following Early-Life Respiratory Syncytial Virus Infection

Summary: This study investigated the effects of low-dose cadmium (Cd) exposure following early-life respiratory syncytial virus (RSV) infection on lung metabolism and inflammation. The results showed that low-dose Cd following early-life RSV infection enhanced lung inflammation and caused metabolic disruption. Histopathology data also indicated the occurrence of fibrosis. This study demonstrates the significant impact of cumulative Cd exposure following early-life RSV infection on subsequent inflammation and lung metabolism.

Cytotoxicity of 2D engineered nanomaterials in pulmonary and corneal epithelium

Summary: Two-dimensional (2D) engineered nanomaterials, widely used in consumer and industrial goods, possess unique chemical and physical characteristics. However, they may cause biological interactions and toxicity towards epithelial cells, especially at first-contact sites such as the eye and lung. This study investigated the cytotoxicity of eight types of 2D nanomaterials and found that hexagonal boron nitride exhibited cytotoxicity in multiple epithelial cell types. Additionally, it did not inhibit wound healing in alveolar epithelial cells at sub-cytotoxic doses. These findings highlight the importance of considering the regional cytotoxicity and cell type variations of nanomaterials before use.
