Organophosphate insecticides disturb neuronal network development and function via non-AChE mediated mechanisms

Lennart V. J. van Melis et al.

Summary: Exposure to organophosphate insecticides has been linked to adverse health effects, particularly neurotoxicity. These insecticides primarily inhibit the activity of acetylcholine esterase, but they can also disrupt calcium homeostasis and have non-AChE-mediated effects. This study assessed the effects of two commonly used organophosphate insecticides and their metabolites on intracellular calcium homeostasis and neuronal function. The results showed that these insecticides inhibit calcium influx and neuronal activity, and the parent compounds were more potent than the metabolites.

FDA-Approved Oximes and Their Significance in Medicinal Chemistry

Jyothi Dhuguru et al.

Summary: Despite scientific advancements, organophosphate poisoning remains a significant threat to human health, causing nearly one million poisoning cases and at least 20,000 deaths annually. Oximes are a crucial class of compounds widely used as antidotes, drugs, and intermediates in the synthesis of pharmacological derivatives. This paper summarizes the FDA-approved oximes, discussing their mechanism of action, pharmacokinetics, and synthesis.

Mechanisms and treatment strategies of organophosphate pesticide induced neurotoxicity in humans: A critical appraisal

Shahid Yousuf Ganie et al.

Summary: Organophosphates (OPs) are commonly used pesticides that can cause neurotoxic effects and other disorders. Humans are exposed to OPs through different routes and biomonitoring of OP markers can help measure and control exposure severity. Standard treatments for OP poisoning include administration of atropine and acetylcholine esterase restoration therapy. It is important to increase awareness about the potential risks of OP exposure and reevaluate exposure levels.

Are the current commercially available oximes capable of reactivating acetylcholinesterase inhibited by the nerve agents of the A-series?

Marcelo C. Santos et al.

Summary: This study reports the design and synthesis of a surrogate for novichok agent, NTMGMP, which can be used for experimental evaluation of antidotes. The efficiency of commercial oximes in reactivating NTMGMP-inhibited acetylcholinesterase was evaluated. Experimental and theoretical studies matched the hierarchy of efficiency and identified trimedoxime as the most promising commercial oxime for reactivation.
