The biosynthetic enzymes that generate o-QMs have great potential for biocatalytic applications, enabling the selective production of desirable compounds that are otherwise difficult to achieve by traditional synthetic methods. Further characterization of these enzymes may lead to the discovery of new enzymes capable of similar chemistry on diverse substrates.

[4+2]-Cycloaddition of 1,1,3,3-tetramethylguanidine and o-quinone methides: synthesis of arene-fused 2-dimethylamino-4H-1,3-oxazines

Summary: A method for synthesizing substituted 4H-1,3-benzoxazines and their fused derivatives bearing N,N-dimethylamino moiety at the 2-position of the 4H-1,3-oxazine ring is developed. The reaction involves the use of phenolic and naphtholic Mannich bases, their iodomethylates or salicylic alcohols, and 1,1,3,3-tetramethylguanidine, and is believed to proceed via the generation of o-quinone methides followed by [4+2]-cycloaddition and elimination of dimethylamine.

Enantioselective Synthesis of Atropisomers via Vinylidene ortho- Quinone Methides (VQMs)

Summary: In this Account, we summarize our recent studies on the enantioselective synthesis of atropisomers using the vinylidene ortho-quinone methides (VQMs) as pluripotent intermediates. The most appealing features of VQMs are the disturbed aromaticity and axial chirality of the allene fragment. We have achieved a variety of axially chiral heterocycles, as well as atropisomeric vinyl arenes and chiral helical compounds, through the use of VQM intermediates. Recently, we successfully isolated and structurally characterized the elusive VQMs, providing direct evidence for their existence and expanding the potential applications of VQM chemistry.

This growth has led to the access of structurally complex heterocyclic frameworks through the synthetic transformations of structurally modified stable p-QMs, focusing on transition-metal, Lewis acid, and base-catalyzed/mediated synthetic transformations to oxygen- and nitrogen-containing heterocycles.
