Association between the Pittsburgh sleep quality index and white matter integrity in healthy adults: a whole-brain magnetic resonance imaging study

Reproducible brain-wide association studies require thousands of individuals

Summary: Magnetic resonance imaging (MRI) has greatly advanced our understanding of the human brain, but similar progress has not been made in mental health research. By analyzing three large neuroimaging datasets, researchers found that brain-wide association studies (BWAS) had smaller effect sizes than previously thought, leading to underpowered studies, inflated effect sizes, and replication failures. As sample sizes increased, replication rates improved and effect size inflation decreased. Functional MRI, cognitive tests, and multivariate methods showed more robust effects in BWAS. Smaller than expected brain-phenotype associations and population subsample variability explain the widespread replication failures in BWAS. Reproducibility in BWAS requires samples with thousands of individuals.

Association of body mass index and its classifications with gray matter volume in individuals with a wide range of body mass index group: A whole-brain magnetic resonance imaging study

Summary: This study examined the association between body mass index (BMI) and brain structure in individuals with a wide range of BMI. The results showed a negative correlation between BMI and cerebellum exterior volume, with lower volumes in the overweight or obese group compared to the underweight or normal group. There were also differences between sexes, with a significant correlation between BMI and cerebellum exterior volume only in women. These findings suggest that abnormal BMI may have an impact on regional gray matter volumes.

Summary: Primary insomnia (PI) is characterized by difficulties in initiating and maintaining sleep along with daytime functioning deficits. DTI studies have shown that patients with PI exhibit white matter disruptions, particularly in frontostriatal, frontothalamic, and corticocortical neural networks, shedding light on the potential etiology and pathology of the disorder. The findings suggest that future research should focus on the neural network alterations in PI patients to better understand the underlying mechanisms of the condition.
