Dichotomous Role of Tumor Necrosis Factor in Pulmonary Barrier Function and Alveolar Fluid Clearance

Rudolf Lucas et al.

Summary: Alveolar-capillary leak is a hallmark of ARDS, which is a potentially lethal complication of severe sepsis, trauma, and pneumonia (including COVID-19). ARDS is characterized by barrier dysfunction, hyper-inflammation, and impaired alveolar fluid clearance, leading to pulmonary permeability edema and compromised gas exchange.

Dexmedetomidine protects against burn-induced intestinal barrier injury via the MLCK/p-MLC signalling pathway

Chao Qin et al.

Summary: The study showed that dexmedetomidine treatment mitigated burn-induced intestinal barrier injury by inhibiting the MLCK/p-MLC signaling pathways, reducing histological damage, enhancing tight junction protein expression, and decreasing inflammation levels.

Postnatal development alters functional compartmentalization of myosin light chain kinase in ovine carotid arteries

Dane W. Sorensen et al.

Summary: The study reveals that the catalytic activity of MLCK is similar in fetal and adult arteries, but the fraction of total MLCK in the functional compartment involved in contraction is significantly greater in fetal arteries. Confocal colocalization analysis showed that MLCK activation was greater in periluminal than periadventitial regions in both age groups.

Telmisartan Mitigates High-Glucose-Induced Injury in Renal Glomerular Endothelial Cells (rGECs) and Albuminuria in Diabetes Mice

Xiaolin Zhan et al.

Summary: This study demonstrated that Telmisartan has protective effects against high glucose-induced injury in human renal glomerular endothelial cells through multiple mechanisms. Telmisartan can improve mitochondrial function, reduce oxidative stress, prevent NADPH oxidase 2 expression, and regulate cell permeability via the MLCK/MLC-2/occludin signaling pathway. Additionally, Telmisartan treatment was shown to ameliorate the reduction of occludin and albuminuria in a diabetic mouse model, suggesting its potential therapeutic role in DN.

Heteromeric TRP Channels in Lung Inflammation

Meryam Zergane et al.

Summary: Activation of TRP channels disrupts endothelial barrier function by increasing permeability, facilitating inflammation and pulmonary edema formation. Heteromeric TRP channel subunits in lung endothelium could be targeted for reducing endothelial permeability as a protective strategy in pulmonary inflammation.

TIGIT modulates sepsis-induced immune dysregulation in mice with preexisting malignancy

Wenxiao Zhang et al.

Summary: TIGIT is an inhibitory receptor that is upregulated in cancer and contributes to impaired antitumor immunity, but its role in sepsis is unclear. Experiments showed that anti-TIGIT antibody reversed T cell apoptosis in cancer septic mice, leading to significant survival benefits for these patients. This suggests that anti-TIGIT therapy may be a potential immunotherapy to improve outcomes in septic cancer patients.
