Mode and dosage time in polyethylene glycol precipitation process influences protein precipitate size and filterability

Maria del Carme Pons Royo et al.

Summary: Precipitation has gained attention as an alternative method for monoclonal antibody purification. We propose a gradual dosage of polyethylene glycol to improve the precipitation process. Our study also investigates the impact of the dosage time and concentration on particle size distribution and the 3D structure of the precipitates.

Effect of module geometry on the sustainable flux during microfiltration of precipitated IgG

Mirko Minervini et al.

Summary: This study examined the effect of membrane module geometry on the sustainable flux and fouling mechanisms in the precipitation-filtration process of monoclonal antibody proteins. The results showed that open channel cassettes had higher sustainable flux compared to hollow fiber modules, which were prone to fiber clogging. These findings provide valuable insights for the design of continuous purification processes for monoclonal antibody products.

Media on-demand: Continuous reconstitution of a chemically defined media directly from solids

Daniel Komuczki et al.

Summary: The study demonstrates that continuous on-demand reconstitution of media results in the same quality of the product compared to traditional batch production, including amino acids, carbohydrates, and osmolality profile. Continuous reconstitution also shows comparable cell cultivation performance on a small and benchtop scale compared to batch reconstitution. Additionally, continuous on-demand media reconstitution significantly reduces the space required for preparation and storage.

End-to-end continuous bioprocessing: Impact on facility design, cost of goods, and cost of development for monoclonal antibodies

Hanna Mahal et al.

Summary: This study presents a systematic approach to evaluate the trade-offs between manufacturing and development costs for mAbs in continuous processing. The analysis showed that single-use continuous facilities offer significant COG savings at annual demands of 100-500 kg, while multiple parallel continuous trains are more advantageous for tonnage demands of 1-3 tons. Additionally, the analysis identified the operational requirements to reach different COG savings in scenarios where batch and continuous facilities offer similar COG.
