Reversing the Epithelial-Mesenchymal Transition in Metastatic Cancer Cells Using CD146-Targeted Black Phosphorus Nanosheets and a Mild Photothermal Treatment

Jinyuan Liu et al.

Summary: This study demonstrates a nanomaterial-based approach to reverse cancer cell EMT and inhibit cell migration. By using engineered black phosphorus nanosheets (BPNSs) and a mild photothermal treatment, the researchers successfully converted highly metastatic breast cancer cells to an epithelial phenotype. The approach targets CD146, a known EMT inducer, and downregulates its expression, disrupting downstream EMT-related signaling pathways. Considering CD146 overexpression in various metastatic cancer cells, this method shows promise for treating cancer metastasis.

Double-drug loading upconversion nanoparticles for monitoring and therapy of a MYC/BCL6-positive double-hit diffuse large B-cell lymphoma

Yulu Yang et al.

Summary: This study discovered that DLBCL genome changes and expression characteristics are associated with various immune cells through whole exome sequencing. Inspired by lenalidomide as an immunomodulatory drug for multiple myeloma, the researchers constructed a multifunctional nanoplatform named UCNPs-Len-Dex, which demonstrated good biocompatibility and antitumor efficacy in vitro and in vivo. The RNA sequencing results indicated that UCNPs-Len-Dex targeted and activated the E3 ligase of CRBN, inhibiting MYC/BCL6-positive DLBCL and maintaining immune microenvironment stability.

Targeting transcription cycles in cancer

Stephin J. Vervoort et al.

Summary: Accurate control of gene expression is crucial for normal development and any dysregulation can lead to cancer initiation and progression. The transcription process can be viewed as a multistep cycle, with dysregulation potentially causing defective gene expression control in cancer. Targeting transcriptional cyclin-dependent kinases and associated proteins may hold promise for cancer treatment by disrupting global or selective transcription.

Application of ZnO/CNT@Fe3O4 nanocomposite in amplifying the anti-leukemic effects of Imatinib: a novel strategy to adjuvant therapy in chronic myeloid leukemia

Amir-Mohammad Yousefi et al.

Summary: The combination of synthesized ZnO/CNT@Fe3O4 nanocomposite with Imatinib showed decreased survival of CML-derived K562 cells through inducing reactive oxygen species-mediated apoptosis. Improved cytotoxicity was found in the presence of a well known autophagy inhibitor, suggesting the enhanced apoptotic effect of this treatment via autophagy suppression. The study also indicated that overexpression of c-Myc plays a contributory role in attenuating the efficacy of ZnO/CNT@Fe3O4-Imatinib in K562 cells.

Transcription Factors: The Fulcrum Between Cell Development and Carcinogenesis

Zeyaul Islam et al.

Summary: Higher eukaryotic development is a complex process regulated by transcription factors, with dysregulation of these factors linked to carcinogenesis. Transcription factors play a crucial role in cancer stemness, supporting the function and maintenance of cancer stem cells. Understanding how these factors interact is crucial for defining cell fate in both embryogenesis and tumor development, with potential implications for developing better research and medical protocols. Dysregulation of TFs is also associated with cancer progression and metastasis, highlighting the potential for TF networks to be targeted therapeutically and specific TFs to serve as biomarkers for treatment responses.
