Role of mTOR through Autophagy in Esophageal Cancer Stemness

Liang Du et al.

Summary: Esophageal cancer is a highly aggressive disease with poor prognosis. The hypoxia-responding mTOR pathway plays a crucial role in the regulation of esophageal cancer stem-like cells (CSCs). Targeting the mTOR pathway could be a potential therapeutic strategy for eliminating putative esophageal CSCs.

Zeb1 modulates hematopoietic stem cell fates required for suppressing acute myeloid leukemia

Alhomidi Almotiri et al.

Summary: The study suggests that loss of Zeb1 in adult stem cells leads to significant defects in stem cell renewal and differentiation, which are associated with leukemia signaling. Therefore, Zeb1 plays a crucial role as a transcriptional regulator in hematopoiesis, coordinating HSC self-renewal and differentiation to suppress the potential development of AML.

KLF4, Slug and EMT in Head and Neck Squamous Cell Carcinoma

Julia Ingruber et al.

Summary: The study found that in head and neck squamous cell carcinoma, Slug gene expression was significantly higher while KLF4 gene expression was significantly lower. There was a significant negative correlation between KLF4 and Slug gene expression in the majority of HNSCC samples. TGF-β1 contributed to the downregulation of KLF4 and the upregulation of Slug.

Hyaluronidase and pH Dual-Responsive Nanoparticles for Targeted Breast Cancer Stem Cells

Weinan Li et al.

Summary: The pH-responsive and CD44 receptor-mediated targeted nanoparticles loaded with Thioridazine showed great potential in eliminating cancer stem cells. In vitro release studies demonstrated effective drug release in acidic pH conditions. Cellular uptake studies confirmed efficient uptake of the nanoparticles in mammospheres, and in vivo distribution studies showed prolonged retention at tumor sites and improved tumor-suppressing effects.

Regulation of Cellular and Cancer Stem Cell-Related Putative Gene Expression of Parental and CD44+CD24- Sorted MDA-MB-231 Cells by Cisplatin

May Zie Koh et al.

Summary: In this study, the attenuated cytotoxicity of cisplatin in CSC-like cells compared to parental cells was observed, along with a distinct set of dysregulated putative target genes related to the Hedgehog pathway and angiogenesis in CSC-like cells after cisplatin treatment. Understanding the role of these dysregulated genes in cisplatin resistance of CSCs may contribute to the development of potential therapeutic targets for cisplatin-resistant breast cancer.

Transcription Factors: The Fulcrum Between Cell Development and Carcinogenesis

Zeyaul Islam et al.

Summary: Higher eukaryotic development is a complex process regulated by transcription factors, with dysregulation of these factors linked to carcinogenesis. Transcription factors play a crucial role in cancer stemness, supporting the function and maintenance of cancer stem cells. Understanding how these factors interact is crucial for defining cell fate in both embryogenesis and tumor development, with potential implications for developing better research and medical protocols. Dysregulation of TFs is also associated with cancer progression and metastasis, highlighting the potential for TF networks to be targeted therapeutically and specific TFs to serve as biomarkers for treatment responses.

Zahra Hosseini-khah et al.

Summary: SOX2 and Bcl-2 are significantly overexpressed in HCC patients, correlating with TNM staging and tumor grading. Patients with SOX2 overexpression have a lower 5-year overall survival rate. There is no significant association between Bcl-2 expression and survival rate.
