The Jun-dependent axon regeneration gene program: Jun promotes regeneration over plasticity

Matthew R. J. Mason et al.

Summary: The regeneration-associated gene expression program is crucial for axon re-growth in injured peripheral neurons. Jun, a transcription factor, plays an important role in regulating this program by upregulating gene expression and promoting cell regeneration while inhibiting plasticity response.

Stimulation Parameters Used During Repetitive Transcranial Magnetic Stimulation for Motor Recovery and Corticospinal Excitability Modulation in SCI: A Scoping Review

Nabila Brihmat et al.

Summary: There is a growing interest in using non-invasive stimulation interventions, such as repetitive transcranial magnetic stimulation (rTMS), to improve functional outcomes and recovery after spinal cord injury (SCI). This scoping review focuses on the stimulation parameters used in 20 rTMS protocols and explores their potential associations with observed effects. Future studies should consider timing, intervention duration, and dosage in relation to the stage, level, and severity of the injury. More real vs. sham rTMS studies with similar designs and replication information are needed to establish a significant level of evidence for the use of rTMS in SCI.

Overlapping transcriptional programs promote survival and axonal regeneration of injured retinal ganglion cells

Anne Jacobi et al.

Summary: This study examined three interventions that enhance survival and regeneration of mouse retinal ganglion cells (RGCs) after optic nerve crush (ONC) injury. Manipulation of multiple pathways is required for type-independent axon regeneration. The analysis of gene expression using single-cell transcriptomics identified genes selectively expressed by RGCs likely to be dying, surviving, or regenerating. Overexpression of genes associated with the regeneration program improved both survival and axon regeneration, suggesting the potential of mechanistic analysis in identifying novel therapeutic strategies.

Yuyan Cheng et al.

Summary: In this study, the authors used integrative genomic analysis and experimental validation to identify REST as a novel suppressor of a pro-regenerative gene program and CNS axon regeneration in mice. They demonstrated the inhibitory role of REST and highlighted the importance of a systems biology approach in CNS repair.

Cellular mechanisms underlying state-dependent neural inhibition with magnetic stimulation

Hui Ye et al.

Summary: Novel stimulation protocols for neuromodulation with magnetic fields have been explored, revealing the significance of the activation state of the nervous system in magnetic stimulation. High frequency magnetic stimulation was found to inhibit neural activity in a state-dependent manner, with the underlying cellular mechanisms related to voltage-dependent ion channels. Monitoring and investigating variability in neural activity is crucial for optimizing the outcomes of magnetic stimulation in research and practice.

Genetic dissection of the glutamatergic neuron system in cerebral cortex

Katherine S. Matho et al.

Summary: The study established genetic strategies and tools to dissect and fate-map subpopulations of pyramidal neurons based on their developmental and molecular programs. Through combinatorial strategies, subsets of pyramidal neurons defined by developmental origin, marker expression, anatomical location, and projection targets can be targeted for viral access. These strategies provide an experimental framework for understanding the hierarchical organization and developmental trajectory of subpopulations of pyramidal neurons.

Molecular Mechanisms Underlying Ascl1-Mediated Astrocyte-to-Neuron Conversion

Zhiping Rao et al.

Summary: Direct neuronal reprogramming through Ascl1 can efficiently convert astrocytes into induced neuronal cells, with key regulatory genes such as Klf10, Myt1, Myt1l, Neurod4, and Chd7 playing important roles in different stages of the conversion process. This study provides valuable insights into the molecular mechanisms underlying Ascl1-mediated astrocyte-to-neuron conversion, enhancing the potential for cell-based therapies.

Article MAPK pathway activation selectively inhibits ASCL1-driven small cell lung cancer

Rebecca Caeser et al.

Summary: Activation of the mitogenic signaling pathway plays a significant role in lung cancer, particularly in small cell lung cancer. Research suggests that the most common subtype of SCLC, SCLC-A, is selectively sensitive to MAPK activation, potentially inhibiting proliferation through inducing cell cycle arrest and senescence.

Paw preferences in mice and rats: Meta-analysis

Martina Manns et al.

Summary: Mice and rats show significant hemispheric asymmetries at the individual level, with a preference for either the left or right paw. However, unlike humans, population level asymmetries were not observed. These results highlight the importance of considering individual hemispheric differences in both basic and clinical neuroscience.

Deletion of Kruppel-like factor-4 promotes axonal regeneration in mammals

Jin-Hui Xu et al.

Summary: The deletion of Klf4 gene enhances axonal regeneration in sensory neurons after peripheral nerve injury, both in vitro and in vivo. Klf4 knockout also promotes corticospinal tract regeneration in mice with spinal cord injury, suggesting Klf4 as a potential target for promoting functional recovery after nervous system damage.
