Fully Automated Optical Hematocrit Measurement from Dried Blood Spots

Marc Luginbuhl et al.

Summary: The impact of hematocrit (HCT) on the spreading area of dried blood spots (DBS) is a major obstacle for quantitative microsampling strategies. Current methods for assessing HCT from DBS are either labor-intensive, require complicated algorithms, or are not automatable. This study presents a novel setup that allows fully automated reflectance analysis to measure HCT from DBS, enabling high-throughput analysis in combination with automated DBS extraction. The setup showed excellent precision and accuracy in analyzing authentic samples and demonstrated the application of HCT correction during a pharmacokinetic study.

PLS-DA vs sparse PLS-DA in food traceability. A case study: Authentication of avocado samples

Ana M. Jimenez-Carvelo et al.

Summary: Conventional PLS-DA and sparse sPLS-DA were effectively used to authenticate avocado samples by analyzing lipid chromatographic fingerprints. The concatenated classification models successfully resolved multiclass problems in food authentication, with performance metrics around 0.95 for both multivariate classification methods.

MetaboAnalyst 5.0: narrowing the gap between raw spectra and functional insights

Zhiqiang Pang et al.

Summary: MetaboAnalyst version 5.0 has been introduced to facilitate comprehensive metabolomics data analysis and interpretation, with the aim of narrowing the gap from raw data to functional insights. Three new modules have been developed to assist in this goal, along with various other new functions and improvements in interface, graphics, and codebase. Users can now easily switch between compatible modules for a streamlined data analysis experience.

Metabolomic Laboratory-Developed Tests: Current Status and Perspectives

Steven Lichtenberg et al.

Summary: Laboratory-developed tests (LDTs) are a subset of in vitro diagnostic devices that are manufactured and used within a single laboratory. This review analyzes the current state and development prospects of LDTs based on metabolomics, comparing them with scientific metabolomics study. Legal aspects of LDT regulation in various regions are discussed, along with recommendations for further introducing metabolomic LDTs into practice.

Optimal Trees Selection for Classification via Out-of-Bag Assessment and Sub-Bagging

Zardad Khan et al.

Summary: The impact of training data size on machine learning methods has been extensively studied, showing that the predictive performance of tree based methods generally improves at a decreasing rate with increasing training data size. In optimal trees ensemble (OTE), modified tree selection methods are proposed to address the issue of failing to learn from some training observations due to internal validation. These modified methods show improved performance compared to OTE and other existing methods, as demonstrated in analysis on benchmark datasets and simulation studies.
