The Eco-Exposome Concept: Supporting an Integrated Assessment of Mixtures of Environmental Chemicals

Stefan Scholz et al.

Summary: The concept of the eco-exposome, analogous to the human exposome, aims to describe the totality of internal exposure over the lifetime of ecologically relevant organisms and explore linking exposure to adverse effects. The study discusses methods and technologies using targeted and nontargeted chemical analyses and bioassays to characterize eco-exposome exposure.

Risk Characterization and Probabilistic Concentration-Response Modeling of Complex Environmental Mixtures Using New Approach Methodologies (NAMs) Data from Organotypic in Vitro Human Stem Cell Assays

Nan-Hung Hsieh et al.

Summary: The study evaluated the validity of traditional mixture risk assessment paradigms IA and CA with NAMs data. Results showed that CA is more accurate in predicting mixture effects compared to IA, but in some cases, mixture bioactivity appeared to be greater than that of individual components.

Summary: The study evaluated chemical-induced changes in gene expression using differentiated HepaRG(TM) cells and a Bayesian framework that quantitatively modeled these changes through six transcription factors. The data provided insights into the molecular signaling network of HepaRG(TM) cell cultures.

OpenCYP: An open source database exploring human variability in activities and frequencies of polymophisms for major cytochrome P-450 isoforms across world populations

Susanna Vichi et al.

Summary: The OpenCYP database provides data on human variability of selected cytochrome P-450 isoforms, with limited data on other age groups. Quantitative comparisons are often hindered by different experimental conditions, but the database is seen as a promising tool for human risk assessment of chemicals.

SCALABLE PENALIZED SPATIOTEMPORAL LAND-USE REGRESSION FOR GROUND-LEVEL NITROGEN DIOXIDE

Kyle P. Messier et al.

Summary: The study developed a scalable approach for simultaneous selection of variables and estimation of LUR models with spatiotemporally correlated errors through a combination of general Vecchia Gaussian-process approximation with a penalty on the LUR coefficients. Compared to existing methods using simulated data, the approach resulted in higher model-selection specificity and sensitivity, as well as better prediction in terms of calibration and sharpness, across a wide range of settings. The spatiotemporal analysis of daily ground-level NO2 data in the United States using this approach was more accurate and produced a sparser and more interpretable model, revealing significant variations between cities and intra-urban variation. The daily predictions can be valuable for epidemiological and risk-assessment studies seeking daily, national-scale predictions and for use in acute-outcome health-risk assessments.

Application of new approach methodologies: ICE tools to support chemical evaluations

Jaleh Abedini et al.

Summary: New approach methodologies (NAMs) are utilized for toxicological assessment, with ICE providing user-friendly access to NAM data and tools for exploring chemical bioactivity and molecular properties. ICE contains curated toxicity testing data and physicochemical property data, along with computationally generated toxicity data and physicochemical parameter predictions.ICE also offers interactive computational tools and the newest tool, Chemical Quest, which allows users to search for structurally similar chemicals within the database.

A method for the analysis of 121 multi-class environmental chemicals in urine by high-performance liquid chromatography-tandem mass spectrometry

Hongkai Zhu et al.

Summary: The study introduces a method for monitoring 121 environmental chemicals in urine samples using solid-phase extraction (SPE) coupled with liquid chromatography-tandem mass spectrometry (LC-MS/MS). The optimized method showed excellent variability, satisfactory recoveries, and high detection limits for a wide range of analytes. This validated method was successfully applied to the simultaneous determination of environmental chemicals in real urine samples.

The Community Multiscale Air Quality (CMAQ) model versions 5.3 and 5.3.1: system updates and evaluation

K. Wyat Appel et al.

Summary: The CMAQ model version 5.3, released in August 2019, features significant updates in science, functionality, and computation efficiency compared to version 5.2.1. Major advances in the new model include enhanced aerosol, chlorine, bromine, and iodine chemistry, as well as updated deposition models. In addition, support for the WRF model's hybrid vertical coordinate was added. Evaluations of CMAQ531 show differences in O-3 and PM2:5 values compared to CMAQ521.
