Optimization of steam explosion parameters for improved biotechnological use of wheat straw

Summary: Using lignocellulosic raw materials as substrate for biotechnological applications has been a focus of research. In this study, the effects of steam explosion combined with water in treating wheat straw were investigated. The results showed that impregnation with water increased the yields of glucose and xylose from subsequent enzymatic hydrolysis. Furthermore, optimizing the steam explosion parameters further increased the maximum saccharide yield. The growth of Yarrowia lipolytica on hydrolysates derived from different steam explosion parameters was also evaluated.

Summary: In this study, the conversion of crude glycerol to citric acid by wild-type yeast Yarrowia lipolytica DSM 8218 was modeled using Response Surface Methodology. The optimal conditions for the conversion were determined and confirmed in a scaled up fermentation experiment, which showed a metabolic shift from mannitol to citric acid when high stirring rates were applied. Additionally, a morphic transition from pseudo-mycelial form to round-shaped yeast-like cells was observed.

Conversion of cardoon crop residues into single cell oils by Lipomyces tetrasporus and Cutaneotrichosporon curvatus: process optimizations to overcome the microbial inhibition of lignocellulosic hydrolysates

Summary: This study investigated the production of Single Cell Oils from undetoxified cardoon hydrolysates, with Lipomyces tetrasporus showing better tolerance to the toxic effects of biomass degradation by-products compared to Cutaneotrichosporon curvatus. In batch mode using SP-inocula and the highest dilution ratio, the maximum lipids yields and lipid cell content were achieved, with oleic acid being the dominant fatty acid. Scaling up to the 2 L bioreactor in fed-batch mode increased lipids concentration and productivity.
