Cutting-edge mass spectrometry strategy based on imaged capillary isoelectric focusing (icIEF) technology for characterizing charge heterogeneity of monoclonal antibody

Summary: Imaging capillary isoelectric focusing (icIEF) technology is the gold standard for monitoring monoclonal antibody (mAb) charge heterogeneity. However, traditional icIEF cannot be used in conjunction with mass spectrometry (MS) and has limitations in characterizing protein heterogeneity in depth. In this study, an integrated icIEF platform was developed that allows for rapid and reliable monitoring of protein heterogeneity, as well as MS coupling and fraction collection for further characterization.

A platform method for charge heterogeneity characterization of fusion proteins by icIEF

Summary: The charge heterogeneity of fusion proteins can vary significantly compared to traditional bio-pharmaceuticals, posing a challenge in their characterization. In this study, a platform method using icIEF was developed to characterize the charge heterogeneity of various fusion protein therapeutics. The method showed reproducible results and comparable peak patterns to the traditional slab gel IEF. It can serve as a starting point for optimization and as an assay for identity and purity of fusion proteins in the bio-pharmaceutical industry.
