Functional activation of the periaqueductal gray matter during conditioned and unconditioned fear in guinea pigs confronted with the Boa constrictor constrictor snake

B. B. de Paula et al.

Summary: This study evaluated the activation pattern of the periaqueductal gray matter (PAG) in guinea pigs during unconditioned and conditioned fear situations using a prey vs predator paradigm. The results showed that the lateral column of the PAG exhibited higher activation of Fos-labeled cells when facing predators, suggesting the recruitment of specific PAG columns may depend on the nature of the threatening stimulus.

Panic-like responses of female Wistar rats confronted by Bothrops alternatus pit vipers, or exposure to acute hypoxia: Effect of oestrous cycle

Renata Ferreira-Sgobbi et al.

Summary: Anxiety-related diseases occur more frequently in women than in men, and symptoms in women may worsen during the late luteal phase of the menstrual cycle. Research on underlying mechanisms and drug development have mainly been conducted using male animals, but a study comparing male and female rats' responses to threatening stimuli found that both sexes exhibit similar defensive behaviors, with females showing exacerbated panic-like behavior during the late dioestrus phase.

Augmented anandamide signalling in the substantia nigra pars reticulata mediates panicolytic-like effects in mice confronted by Crotalus durissus terrificus pit vipers

Rafael C. Almada et al.

Summary: The enhancement of endogenous AEA in the SNpr through CB1 receptor signaling has been shown to have panicolytic effects, reducing defensive behaviors in mice confronted with rattlesnakes.

CB1 receptor signalling mediates cannabidiol-induced panicolytic-like effects and defensive antinociception impairment in mice threatened by Bothrops jararaca lancehead pit vipers

Bruno Mangili de Paula Rodrigues et al.

Summary: This study investigated the effect of cannabidiol (CBD) on panic-like defensive responses in mice threatened by the venomous snake Bothrops jararaca. The results showed that CBD significantly reduced risk assessment and escape responses in mice. The study suggests that the anti-aversive effect of CBD depends at least partly on the recruitment of CB1 receptors.

Nitric oxide-mediated defensive and antinociceptive responses organised at the anterior hypothalamus of mice are modulated by glutamatergic inputs from area 24b of the cingulate cortex

Luiz Luciano Falconi-Sobrinho et al.

Summary: The study found that activation of N-methyl-D-aspartic acid receptors in Cg1 area increased escape behavior, reduced freezing behavior, and attenuated antinociceptive responses to SIN-1 microinjections in the anterior hypothalamus. Neurotracing and immunohistochemistry results suggest that these responses are mediated by glutamatergic synapses at the anterior hypothalamus.

The modulation of striatonigral and nigrotectal pathways by CB1 signalling in the substantia nigra pars reticulata regulates panic elicited in mice by urutu-cruzeiro lancehead pit vipers

Rafael Carvalho Almada et al.

Summary: This study investigated the role of the cannabinoid receptor type 1 (CB1R) in the substantia nigra pars reticulata (SNpr) in modulating fear responses in mice facing pit vipers. The results indicate that CB1R in the SNpr may regulate the defensive behaviors of mice in threatening situations by modulating the activity of GABAergic neurons from the striatum in this region.

Dorsal premammillary projection to periaqueductal gray controls escape vigor from innate and conditioned threats

Weisheng Wang et al.

Summary: The study showed that PMd-cck cells in mice are activated during escape, and inhibition of the PMd decreases escape speed. The PMd-dlPAG circuit is identified as a central node controlling escape behavior in response to threats.
