The Parallel Reaction Monitoring-Parallel Accumulation-Serial Fragmentation (prm-PASEF) Approach for Multiplexed Absolute Quantitation of Proteins in Human Plasma

Alexander Brzhozovskiy et al.

Summary: Mass spectrometry-based quantitative proteomic methods, such as prm-PASEF, are important tools for protein biomarker discovery and validation. This study demonstrated the use of prm-PASEF for absolute protein quantitation in human plasma, with improved sensitivity and a broad dynamic range. The prm-PASEF method showed excellent correlation with MRM, highlighting its suitability for absolute quantitative proteomics.

Metabolomics based plasma biomarkers for diagnosis of oral squamous cell carcinoma and oral erosive lichen planus

Xibo Li et al.

Summary: This study identified biomarkers associated with oral squamous cell carcinoma (OSCC) and oral erosive lichen planus (OELP) by analyzing plasma samples. The biomarkers were found to be closely related to cholic acid metabolism and amino acid metabolism. By establishing a diagnostic panel, it was possible to predict the malignant transformation of OELP and diagnose OSCC in its early stages.

Highly Multiplexed Targeted Proteomics Acquisition on a TIMS-QTOF

Antoine Lesur et al.

Summary: Targeted proteomics with high sensitivity is achieved through the detection of specific peptides and proteins in complex biological samples. By utilizing the prm-PASEF method with trapped ion mobility separation, over 200 peptides can be monitored in a 30-minute liquid chromatography separation, improving quantification accuracy in biological samples. Detection and quantification of isotope-labeled synthetic peptides with limits of quantification as low as 17.2 amol were achieved, demonstrating the reproducibility and accuracy of the method.

Protein Cargo of Salivary Small Extracellular Vesicles as Potential Functional Signature of Oral Squamous Cell Carcinoma

Simona Fontana et al.

Summary: This pilot study utilized proteomic and bioinformatic strategies to analyze saliva small extracellular vesicles (S/SEVs) from oral squamous cell carcinoma (OSCC) patients, identifying 365 proteins that could potentially serve as novel biomarkers for early OSCC diagnosis. The analysis also revealed specific cluster of enriched functional network terms associated with each group of S/SEVs, indicating their potential value in predicting OSCC.

Plasma-Derived Exosomal microRNA-130a Serves as a Noninvasive Biomarker for Diagnosis and Prognosis of Oral Squamous Cell Carcinoma

Tao He et al.

Summary: Exosomal miR-130a was found to be significantly upregulated in oral squamous cell carcinoma (OSCC) patients compared to healthy controls, and showed correlations with advanced tumor stage, TNM stage, and poorly differentiated OSCC. High levels of exosomal miR-130a were associated with poorer 3-year overall survival and recurrence-free survival in OSCC patients, indicating its potential as a diagnostic and prognostic biomarker.
