THEM6-mediated reprogramming of lipid metabolism supports treatment resistance in prostate cancer

Arnaud Blomme et al.

Summary: THEM6 has been identified as a marker of ADT resistance in PCa, with its deletion reducing tumor growth and restoring castration sensitivity. It regulates intracellular levels of ether lipids to induce the ER stress response, altering ER function and preventing lipid-mediated activation. High THEM6 expression is associated with poor survival and high levels of UPR activation in PCa patients.

Intercepting IRE1 kinase-FMRP signaling prevents atherosclerosis progression

Zehra Yildirim et al.

Summary: ER stress induces phosphorylation of FMRP, leading to suppression of cholesterol efflux and efferocytosis. FMRP deficiency and inhibition of IRE1 kinase activity enhance cholesterol efflux and efferocytosis, reducing atherosclerosis.

Neurotherapeutic implications of sense and respond strategies generated by astrocytes and astrocytic tumours to combat pH mechanical stress

Sebastian John et al.

Summary: This study found that low extracellular pH is perceived as mechanical stress by cells. GM3 acts as a lipid-based pH sensor, generating plasma membrane deformation stress in low pH. This leads to the activation of the IRE1-sXBP1-SREBP2-ACSS2 response axis, promoting cholesterol biosynthesis and surface trafficking to prevent acid-mediated membrane hydrolysis and cell death.

Induction and Aggravation of the Endoplasmic-Reticulum Stress by Membrane-Lipid Metabolic Intermediate Phosphatidyl-N-Monomethylethanolamine

Yuki Ishiwata-Kimata et al.

Summary: This article investigates the synthesis pathways of phosphatidylcholine (PC) and its role in endoplasmic reticulum (ER) stress. The study found that the opi3 gene mutation can induce ER stress even when free choline is sufficiently supplied. Additionally, monomethylethanolamine produced through the Kennedy pathway may act as a molecule that induces ER stress.

Destabilization of β Cell FIT2 by saturated fatty acids alter lipid droplet numbers and contribute to ER stress and diabetes

Xiaofeng Zheng et al.

Summary: Western-type diets, high in saturated fatty acids (SFAs), are associated with obesity and diabetes. This study reveals that SFAs, but not unsaturated fatty acids (USFAs), reduce lipid droplets (LDs) in pancreatic beta cells. SFAs induce the degradation of fat storage-inducing transmembrane protein 2 (FIT2), inhibiting LD formation. Reduction of FIT2 in beta cells impairs cellular functions and exacerbates diet-induced diabetes.

Dietary cholesterol drives fatty liver-associated liver cancer by modulating gut microbiota and metabolites

Xiang Zhang et al.

Summary: High dietary cholesterol leads to sequential progression of steatosis, steatohepatitis, fibrosis and eventually HCC in mice, concomitant with insulin resistance. Cholesterol-induced NAFLD-HCC formation is associated with gut microbiota dysbiosis, with specific bacterial composition changes between stages of liver disease. Alterations in gut bacterial metabolites were also observed with dietary cholesterol intake. Therapies targeting cholesterol and gut microbiota may be effective for preventing NAFLD-HCC.

IRE1α RIDD activity induced under ER stress drives neuronal death by the degradation of 14-3-3 θ mRNA in cortical neurons during glucose deprivation

Juan Carlos Gomora-Garcia et al.

Summary: The study explores the relationship between altered protein homeostasis and neurodegenerative diseases, finding that glucose deprivation triggers the activation of the PERK and IRE1 alpha branches of the UPR, leading to neuronal death.

Regulation of lipid metabolism by the unfolded protein response

Matthieu Moncan et al.

Summary: The endoplasmic reticulum is responsible for protein folding, secretion, Ca2+ storage, and lipid synthesis in eukaryotic cells. Disruption to protein folding or Ca2+ homeostasis in the ER leads to what is known as ER stress, triggering the unfolded protein response pathway. Three ER membrane proteins play a crucial role in this process, sensing the accumulation of unfolded/misfolded proteins and activating an integrated transcriptional program.

SREBP1/FASN/cholesterol axis facilitates radioresistance in colorectal cancer

Yuxiao Jin et al.

Summary: Cholesterol enhances radioresistance of CRC cells by promoting cell proliferation and inhibiting apoptosis through the SREBP1/FASN signaling pathway. Blocking this pathway accelerates radiation-induced CRC cell death, suggesting a potential target for CRC treatment. Lowering cholesterol intake may benefit CRC patients undergoing radiotherapy.

Mechanisms of stearoyl CoA desaturase inhibitor sensitivity and acquired resistance in cancer

Nicole Oatman et al.

Summary: In glioblastoma, some patients have low levels of SCD expression, leading to the evolution of cell lines that can survive independent of SCD, despite unknown mechanisms. Cell lines that escape genetic and epigenetic alterations become highly dependent on SCD for survival. Resistance to SCD inhibition in SCD-dependent lines is mediated through a previously unknown FOSB mechanism, which can be blunted to extend survival in tumor-bearing mice.

Phosphatidylcholine mediates the crosstalk between LET-607 and DAF-16 stress response pathways

Bin He et al.

Summary: This study reveals a crosstalk between stress response pathways in C. elegans, indicating that suppression of LET-607 adaptively activates DAF-16, extending worm lifespan; meanwhile, sphingomyelin synthase SMS-5 plays a critical role in this communication, activating DAF-16 by reducing specific membrane lipid PC levels.

These results highlight the crucial role of IEC phospholipid equilibrium in colonic homeostasis.

Lipidomic atlas of mammalian cell membranes reveals hierarchical variation induced by culture conditions, subcellular membranes, and cell lineages

Summary: Lipid membranes are essential for the structural and functional compartmentalization of cells and sub-cellular organelles. This study used shotgun lipidomics to analyze the lipidomes of various cultured and primary mammalian membrane preparations, identifying clear differences between cultured and primary cells, as well as distinct lipidomic signatures of individual cell lines and tissue types. These findings contribute to a comprehensive understanding of mammalian lipidomes and their regulation in various contexts.
