A genotyping assay to determine geographic origin and transmission potential of Plasmodium falciparum malaria cases

Summary: The study analyzed the Plasmodium falciparum Pfs47 gene sequences and identified two SNP loci that can determine the likely continent of origin of P. falciparum isolates. Sequences from Africa, Asia, and the New World showed over 99% frequency of unique combinations of the SNPs 707 and 725 genotypes, while Papua New Guinea sequences exhibited the highest diversity in these SNPs. Development of accurate High-Resolution Melting assays for genotyping these SNPs can help assess the geographic origin and transmission risk of imported P. falciparum malaria cases.
