HLA-independent T cell receptors for targeting tumors with low antigen density

Jorge Mansilla-Soto et al.

Summary: HLA-independent T cell receptors (HIT receptors) are a novel type of chimeric antigen receptors that can effectively target tumors with low antigen expression, surpassing the sensitivity of CD28-based receptors. They offer high antigen sensitivity and can recognize tumors beyond the capabilities of other CAR designs. The functional persistence of HIT T cells can be enhanced by coexpression of CD80 and 4-1BBL. HIT receptors have been validated in xenograft mouse models, targeting CD19 and CD70 in B cell leukemia and acute myeloid leukemia, respectively.

CAR T cell killing requires the IFN gamma R pathway in solid but not liquid tumours

Rebecca C. Larson et al.

Summary: CAR T cell therapy has shown limited efficacy against solid tumours, and this study identifies the loss of genes in the interferon-gamma receptor signalling pathway as a potential resistance mechanism in glioblastoma and other solid tumours. The study reveals that the interferon-gamma receptor signalling pathway is critical for the adhesion and cytotoxicity of CAR T cells in solid tumours, highlighting the importance of enhancing binding interactions between T cells and tumour cells for improved responses in solid tumours.

Size-dependent activation of CAR-T cells

Qian Xiao et al.

Summary: The activation of CAR-T cells is dependent on the size difference between the CAR-antigen pair and CD45. Increasing the size of CAR extracellular domains weakens CAR-T activation, while membrane-proximal epitopes activate CAR-Ts better than membrane-distal epitopes.

Extensive functional comparisons between chimeric antigen receptors and T cell receptors highlight fundamental similarities

Xueyin Wang et al.

Summary: The study conducted a large-scale, systematic comparison of CARs and TCRs across 5 different pMHC targets and found no consistent difference between CARs and TCRs in terms of their sensitivity to major regulators of T cell activation. While TCRs often emerge as potent, selective receptors directly from human blood, CARs need to be heavily optimized to attain these properties.

Quantitative contributions of TNF receptor superfamily members to CD8+ T-cell responses

John Nguyen et al.

Summary: The study found that among co-stimulation TNF receptor superfamily members, 4-1BB and CD27 increased cytokine production in CD8(+) T cells, but only 4-1BB could prolong the duration of cytokine production. CD27 co-stimulation could enhance the expression of 4-1BB and GITR, leading to synergy between co-stimulation receptors.

T cell circuits that sense antigen density with an ultrasensitive threshold

Rogelio A. Hernandez-Lopez et al.

Summary: The study engineered a two-step positive-feedback circuit that enables human cytotoxic T cells to discriminate targets based on a sigmoidal antigen-density threshold. This circuit involves a low-affinity synthetic Notch receptor controlling the expression of a high-affinity CAR, leading to sharp discrimination between cancer cells and normal cells based on antigen density.

Chimeric STAR receptors using TCR machinery mediate robust responses against solid tumors

Yue Liu et al.

Summary: A new double-chain chimeric receptor STAR was constructed to mediate strong and sensitive TCR-like signaling upon antigen stimulation, showing better antigen sensitivity and less risk of dysfunction compared to traditional CAR-T cells. In multiple solid tumor models, STAR-T cells outperformed BBzCAR-T cells and showed superior antitumor effects.

Comparative analysis of TCR and CAR signaling informs CAR designs with superior antigen sensitivity and in vivo function

Alexander Salter et al.

Summary: CAR-modified T cell therapy is effective in treating certain hematological malignancies, but requires enhancements to target solid tumors with low antigen density. By examining T cell signaling, researchers were able to design CARs with improved antigen sensitivity, leading to enhanced efficacy in low antigen density environments.

The discriminatory power of the T cell receptor

Johannes Pettmann et al.

Summary: The study found that while TCR discrimination is enhanced compared to conventional cell-surface receptors, it is not perfect. Human T cells can respond to pMHC with affinities as low as K-D around 1 mM, and the kinetic proofreading mechanism explains this extraordinary sensitivity.

Perfect adaptation of CD8+ T cell responses to constant antigen input over a wide range of affinities is overcome by costimulation

Nicola Trendel et al.

Summary: Maintaining and limiting T cell responses to constant antigen stimulation is crucial for controlling pathogens and self-tolerance. Research shows that T cells can exhibit perfect adaptation with partial TCR down-regulation, which can be achieved through a mechanistic model. These findings highlight that perfect adaptation limits T cell responses, making them dependent on costimulation for sustained responses.
