Patients' choices and opinions on chorionic villous sampling and non-invasive alternatives for prenatal testing following preimplantation genetic testing for hereditary disorders: A cross-sectional questionnaire study

Christian L. F. Toft et al.

Summary: This study investigated the choices and reasoning behind chorionic villous sampling and opinions on non-invasive prenatal testing among women and men who achieved pregnancy following preimplantation genetic testing (PGT) for hereditary disorders. The results showed that approximately half of the patients declined chorionic villous sampling, primarily due to the perceived risk of miscarriage. If offered, nine out of ten patients stated they would have chosen non-invasive prenatal testing. Some patients were not aware that the nuchal translucency scan rarely provides information on the hereditary disorder for which PGT was performed. Improving counseling on available prenatal tests and screenings may assist patients in making better informed decisions. Non-invasive prenatal testing is welcomed by the patients and is likely to increase the number of patients opting for confirmatory prenatal testing following PGT for hereditary disorders.

A novel method for noninvasive diagnosis of monogenic diseases from circulating fetal cells

Liang Chang et al.

Summary: A method for noninvasive prenatal testing of monogenic diseases by isolating fetal cells from maternal blood and using direct sequencing and targeted NGS-based SNP haplotyping was successfully established. The fetal origin of the cells and the identification of pathogenic mutations were confirmed through genetic analysis, showing consistent results with amniotic fluid samples. This single cell haplotyping method can be used for diagnosing various genetic diseases.

Use of amplicon-based sequencing for testing fetal identity and monogenic traits with Single Circulating Trophoblast (SCT) as one form of cell-based NIPT

Xinming Zhuo et al.

Summary: This study aimed to develop a rapid, robust, versatile, and low-cost next-generation sequencing method for cell-based non-invasive prenatal testing (NIPT) to distinguish individual fetal cells from maternal cells. By amplifying highly polymorphic genomic regions and utilizing specific SNPs, the method showed potential for single-cell genotyping and identification of disease-causing variants in genetic disorders.

Cell-based non-invasive prenatal diagnosis in a pregnancy at risk of cystic fibrosis

Line Dahl Jeppesen et al.

Summary: Cell-based NIPT accurately determined fetal variant status and differentiated fetal trophoblasts from maternal cells, suggesting a potential less invasive alternative to chorionic villous sampling in the future.
