The human O-GlcNAcome database and meta-analysis

Additionally, a comprehensive meta-analysis of human O-GlcNAcylated proteins has revealed that O-GlcNAcylation is a master regulator of cell signaling.

Danggui-Shaoyao-San improves cognitive impairment through inhibiting O-GlcNAc-modification of estrogen α receptor in female db/db mice

Summary: The study aimed to investigate the neuroprotective effects of Danggui-Shaoyao-San (DSS) on db/db mice with diabetes-induced cognitive dysfunction. Results showed that DSS treatment improved learning and memory abilities, relieved hippocampal neuronal damage, enhanced glucose tolerance and insulin resistance, reduced hyperlipemia, and increased antioxidant enzyme activities while decreasing inflammatory mediator levels. Additionally, DSS upregulated neurotrophic factors and decreased apoptosis in db/db mice.

Pancreatic β-Cell O-GlcNAc Transferase Overexpression Increases Susceptibility to Metabolic Stressors in Female Mice

Summary: The study revealed that beta-cell Ogt overexpression in mice led to glucose intolerance under metabolic stressors such as high-fat diet and streptozocin, despite normal insulin secretion and beta-cell mass. Additionally, female mice with beta Ogt overexpression developed hyperglycemia and glucose intolerance post-treatment with streptozocin, contrary to the typical resistance seen in female mice. Transcriptional analysis showed common altered pathways involving pro-survival and inflammatory regulators in islets with loss or gain of Ogt. These findings suggest a gene dosage effect of Ogt and highlight the importance of O-GlcNAc cycling in beta-cell survival and function for glucose homeostasis.
