Targeting PDE4 as a promising therapeutic strategy in chronic ulcerative colitis through modulating mucosal homeostasis

Heng Li et al.

Summary: Inhibition of PDE4 has been found to be an effective strategy for treating chronic ulcerative colitis (UC) by modulating the cAMP pathway and improving clinical symptoms. This study introduces apremilast as a PDE4 inhibitor and demonstrates its therapeutic capacity in a murine model of chronic UC. It shows that apremilast can restore mucosal homeostasis and improve intestinal barrier function, as well as regulate gut microbiota and antimicrobial responses.

The influence of cytokines on the complex pathology of ulcerative colitis

Hiroshi Nakase et al.

Summary: Ulcerative colitis (UC) is specifically affected by genetic alterations, environmental factors, microbiota, and mucosal immune dysregulation, leading to a lack of response in 30%-55% of patients to current therapies. The expression levels of cytokines associated with treatment depend on disease stage and patient characteristics, suggesting that future targeted therapies should focus on these differences and individual cytokine profiles.

Natural compounds as safe therapeutic options for ulcerative colitis

Mukta Gupta et al.

Summary: Ulcerative colitis (UC) is a chronic inflammatory bowel disease with unknown cause. Conventional treatments for UC have limitations and side effects. Natural remedies, with comparatively fewer side effects, have been used for centuries. This review explores the role of herbal drugs, such as gymnemic acid, shagoal, catechin, curcumin, arctigenin, and boswellic acid, in the treatment of UC. Further research is needed to establish the clinical usefulness and molecular mechanisms of these natural products.

Hypericum sampsonii attenuates inflammation in mice with ulcerative colitis via regulation of PDE4/PKA/CREB signaling pathway

Yinsi Lin et al.

Summary: This study investigates the therapeutic effects and molecular mechanisms of Hypericum sampsonii in a mouse model of ulcerative colitis. The results demonstrate that Hypericum sampsonii can alleviate ulcerative colitis by improving inflammation, protecting intestinal barrier function, inhibiting PDE4, and activating the PKA/CREB signaling pathway.

Targeting IL-10, ZO-1 gene expression and IL-6/STAT-3 trans-signaling by a combination of atorvastatin and mesalazine to enhance anti-inflammatory effects and attenuates progression of oxazolone-induced colitis

Nageh Ahmed El-Mahdy et al.

Summary: The combination therapy of mesalazine and atorvastatin in a rat model of oxazolone colitis significantly attenuated the progression of ulcerative colitis by reducing symptoms like diarrhea, rectal bleeding, and weight loss, while increasing levels of anti-inflammatory factors. This combination treatment showed superior effects compared to single drug therapy.

NAIL: an evolutionarily conserved lncRNA essential for licensing coordinated activation of p38 and NFκB in colitis

Semih Can Akincilar et al.

Summary: By identifying the NF kappa B-specific lncRNA NAIL, this study reveals its role in regulating the activation of NF kappa B and p38 pathways by sequestering and inactivating Wip1, ultimately leading to the initiation and progression of colitis. Its expression highly correlates with NF kappa B activity, making it a potential biomarker and therapeutic target for IBD and other inflammation-associated diseases.

Apple polyphenols extract alleviated dextran sulfate sodium-induced ulcerative colitis in C57BL/6 male mice by restoring bile acid metabolism disorder and gut microbiota dysbiosis

Fang Liu et al.

Summary: The study revealed that both apple polyphenols extract (APE) and phloretin have significant preventive effects on DSS-induced ulcerative colitis (UC), improving UC by inhibiting body weight loss, preventing colon shortening, and mucosa damage. While both APE and phloretin shared some common mechanisms in molecular pathways, they also exhibited some differences in their effects on certain bio markers.

Inhibition of Phosphodiesterase-4 in Psoriatic Arthritis and Inflammatory Bowel Diseases

Andrea Picchianti-Diamanti et al.

Summary: Phosphodiesterases (PDEs) are a diverse family of enzymes that regulate intracellular second messengers, with PDE4 being the most studied isoenzyme known for its role in down-regulating inflammatory responses. Inhibition of PDE4 can increase levels of cAMP, leading to decreased expression of inflammatory factors and increased regulatory cytokines, making it a potential therapeutic target for chronic inflammatory conditions.

Dioscin ameliorates murine ulcerative colitis by regulating macrophage polarization

Mei-Mei Wu et al.

Summary: Restoring immune balance by targeting macrophage polarization with Dioscin is a promising therapeutic strategy for UC. Dioscin ameliorates colitis in mice by inhibiting M1 and promoting M2 macrophage polarization through modulation of mTOR signals. This protection is achieved by inhibiting aerobic glycolysis and promoting fatty acid oxidation in macrophages.
