Detection and quantification of the vacuolar H+ATPase using the Legionella effector protein SidK

Michelle E. Maxson et al.

Summary: This study shows that proton-pumping vacuolar H+ ATPases (V-ATPases) are responsible for the acidic environment in secretory and endocytic organelles. The researchers developed a new probe derived from SidK protein to visualize and quantify V-ATPases with high specificity. They found that the density of V-ATPases increases during phagosome maturation and their distribution varies in lysosomes depending on their subcellular localization.

TRPMLs and TPCs: Targets for lysosomal storage and neurodegenerative disease therapy?

Einar Krogsaeter et al.

Summary: Neurodegenerative diseases (ND) pose a serious health burden and are characterized by the accumulation of damaged proteins. Lysosomes, responsible for protein clearance, play a crucial role in neurodegeneration. Lysosomal storage disorders (LSDs) are severe neurodegenerative diseases caused by mutations in lysosomal enzymes or membrane proteins. Defects in intracellular transport pathways involving endosomes and lysosomes contribute to the pathology of ND.

Defining mitochondrial protein functions through deep multiomic profiling

Jarred W. Rensvold et al.

Summary: Mitochondria, as the epicentres of eukaryotic metabolism and bioenergetics, are linked to numerous diseases. However, the functions of many mitochondrial proteins are still unclear, and the genetic basis of many mitochondrial disorders remains unresolved. Through mass spectrometry-based multiomics analyses, we have established a comprehensive functional compendium of human mitochondrial proteins, providing a foundation for mechanistic investigations and genetic diagnosis of mitochondrial diseases. Our findings have identified specific proteins associated with mitochondrial function and disorders, and defined a rich cell signature of mitochondrial dysfunction.

Parkinson's disease-risk protein TMEM175 is a proton-activated proton channel in lysosomes

Meiqin Hu et al.

Summary: A genetic risk factor for Parkinson's disease, TMEM175, acts as a proton-activated, proton-selective channel on the lysosomal membrane to regulate lysosomal H+ leak. Activation of TMEM175 stops further acidification of lysosomes beyond the normal range. Deficiency of TMEM175 causes lysosomal over-acidification, impaired proteolytic activity, and facilitates alpha-synuclein aggregation.
