Neuroprotective effect of histamine H3 receptor blockade on methamphetamine-induced cognitive impairment in mice

Histamine H3R antagonist counteracts the impaired hippocampal neurogenesis in Lipopolysaccharide-induced neuroinflammation

Summary: This study found that inhibition of H3R reduced microglia activity and promoted a shift from pro-inflammatory M1 phenotype to anti-inflammatory M2 phenotype in microglia, thereby attenuating lipopolysaccharide-induced neuroinflammation in mice. Additionally, inhibition of H3R rescued the impairments of neurogenesis and cognitive function caused by neuroinflammation. Mechanistically, the neuroprotective effects of H3R inhibition were mediated by histamine-dependent activation of H2R and reinforcement of CREB-CBP interaction, which led to increased release of anti-inflammatory cytokines and brain-derived neurotrophic factor, as well as decreased release of pro-inflammatory cytokines.

Summary: Recent evidence suggests that the expression levels of histamine receptor H3 (Hrh3) are upregulated in various cancers, including non-small cell lung cancer (NSCLC). High levels of Hrh3 are associated with poor overall survival in NSCLC patients. Inhibition of Hrh3 has shown to inhibit cell growth, induce apoptosis, and suppress migration and invasion of NSCLC cells through signaling pathways such as PI3K/Akt/mTOR and MEK/ERK. Furthermore, in animal studies, Hrh3 antagonist CPX was able to significantly inhibit tumor growth and modulate expression of EMT markers in NSCLC xenograft models.
