Summary: Traumatic injury of the central nervous system, such as traumatic brain injury (TBI) and spinal cord injury (SCI), results in cell loss and axonal degeneration, but limited regeneration. Stem cell therapies show promise in experimental CNS trauma, but the mechanisms behind their effects are unclear. This review discusses the pros and cons of transplantation methods and recruitment of endogenous stem cells, and proposes potential successful treatment strategies.

Advances in Acute Ischemic Stroke Therapy

Summary: The treatment of acute ischemic stroke is advancing with intravenous thrombolysis, mechanical thrombectomy, and cytoprotection being investigated. Later treatment with alteplase and the use of tenecteplase are beneficial alternatives to alteplase. Cytoprotection as an adjunct to reperfusion has shown no efficacy in prior clinical trials, but may have potential benefits.

Interleukin-4 receptor signaling modulates neuronal network activity

Summary: Evidence suggests that immune responses are not only involved in diseases in the central nervous system (CNS), but also play a crucial role in maintaining healthy synaptic function. The IL-4/IL-4R alpha signaling pathway was found to be important in synaptic processes, with IL-4R alpha expressed presynaptically and regulating synaptic transmission. Deficiency of IL-4R alpha led to altered synaptic function and increased network activity in vivo.

The Role of Interleukin-4 and Interleukin-10 in Osteoarthritic Joint Disease: A Systematic Narrative Review

Summary: This review examines the potential and mechanisms of interleukin-4 (IL-4), interleukin-10 (IL-10), and the IL-4/IL-10 fusion protein (IL4-10 FP) for the treatment of osteoarthritis. The findings suggest that IL-4 and IL-10 have protective effects on osteoarthritic cartilage and can reduce inflammation. IL4-10 FP, as a combination of both cytokines, shows synergistic effects by counteracting proinflammatory effects. Additionally, IL-4 and IL4-10 FP have analgesic properties.

Close liaison with an ophthalmologist is recommended for those patients with a history of severe ocular surface disease or who develop conjunctivitis.

Gene-based therapies for neurodegenerative diseases

Advances in viral vectors and genome-engineering tools have opened up new possibilities for diffusely delivering genes throughout the CNS and manipulating disease pathways, paving the way for a future where gene-based therapeutics are integrated into clinical practice.

This metabolic shift is important for minimizing cytotoxic responses of microglia and macrophages.
