Microbiome Profile and Molecular Pathways Alterations in Gastrointestinal Tract by Hydrogen Sulfide-Releasing Nonsteroidal Anti-Inflammatory Drug (ATB-352): Insight into Possible Safer Polypharmacy

Urszula Glowacka et al.

Summary: The study showed that ATB-352 has better gastrointestinal safety compared to ketoprofen after long-term treatment, maintaining gastrointestinal function by releasing H2S. Compared to ketoprofen, ATB-352 has a smaller impact on intestinal microbiome and molecular sensors and effectively inhibits cyclooxygenase activity.

Intestinal butyrate-metabolizing species contribute to autoantibody production and bone erosion in rheumatoid arthritis

Jing He et al.

Summary: This study demonstrates that raffino-oligosaccharide (ROS) can prevent constipation and improve gastrointestinal transit rate and defecation frequency in mice. It also regulates the diversity and structure of intestinal flora. Targeted screening reveals 29 targets related to the anti-constipation effects of ROS, and network pharmacology analysis identifies several core anti-constipation targets.

Dose- and Sex-Dependent Bidirectional Relationship between Intravenous Fentanyl Self-Administration and Gut Microbiota

Michelle Ren et al.

Summary: Gut bacteria play a role in addiction-related behaviors by influencing neural circuits. The interaction between gut bacteria and the brain is hypothesized to mediate the rewarding and reinforcing properties of fentanyl. Factors such as sex, dose, and gut microbiota impact fentanyl intake. The diversity of gut microbiota predicts fentanyl self-administration, with potential implications for addiction interventions.

Summary: The gut microbiota plays a crucial role in immune regulation and the development of autoimmune diseases such as rheumatoid arthritis (RA), with research into this area being a recent focus. Multiple studies have shown alterations in the composition and/or function of gut microbiota in early and established RA patients, but the molecular mechanisms behind how gut microorganisms induce immune tolerance loss or contribute to disease progression are still poorly understood.

Prenatal opioid administration induces shared alterations to the maternal and offspring gut microbiome: A preliminary analysis

Gregory G. Grecco et al.

Summary: This study used a mouse model to investigate the effects of prenatal methadone exposure on the microbiome of methadone-treated dams and their offspring. Results showed that methadone treatment significantly increased microbial diversity and abundance, with a strong positive correlation between dam and offspring measures of diversity. These preliminary findings suggest that maternal opioid treatment during pregnancy alters the composition of the maternal microbiome, which is also observed in offspring and may contribute to impaired developmental phenotypes.

Zhitong Deng et al.

Summary: Slow transit constipation (STC) is a prevalent medical issue, with lactulose and Chinese Herb Solid Drink (CHSD) both showing potential in relieving constipation by impacting the gut microbiota through different mechanisms. While lactulose acts as a bulking laxative and microbioecologic, CHSD contains medicine food homologous materials which enhance the relative abundance of specific gut microbiota. Both CHSD and lactulose elevate short-chain fatty acids and neurotransmitters, promoting intestinal peristalsis, although they have distinct effects on certain gut neuroendocrine factors like gastrin expression.
