Prediction of Maternal and Fetoplacental Concentrations of Cefazolin, Cefuroxime, and Amoxicillin during Pregnancy Using Bottom-Up Physiologically Based Pharmacokinetic Models

Khaled Abduljalil et al.

Summary: This study developed a PBPK model to predict the disposition of certain drugs in maternal and fetal tissues. The model accurately predicted the exposure levels in both the mother and fetus, and could be used for clinical trial design and risk assessment for maternal-fetal exposure.

Re-writing Oral Pharmacokinetics Using Physiologically Based Finite Time Pharmacokinetic (PBFTPK) Models

Pavlos Chryssafidis et al.

Summary: The purpose of this study was to develop physiologically based finite time pharmacokinetic models for the analysis of oral pharmacokinetic data. The models were based on the passive drug diffusion mechanism under the sink conditions principle. Simulated data and experimental literature data were used to validate the models. The results showed that the PBFTPK models are a powerful tool for analyzing oral pharmacokinetic data.

Columbus' egg: Oral drugs are absorbed in finite time

P. Macheras et al.

Summary: The assumption of infinite time for oral drug absorption has had a significant impact on the study of bioavailability and bioequivalence. However, it has been revealed that drug absorption has a finite time period, calling for regulatory changes in assessment methods.

A Signal Processing Approach to Pharmacokinetic Data Analysis

C. O. S. Sorzano et al.

Summary: This article examines the connection between pharmacokinetic models and system theory, demonstrating how system theory can easily accommodate different therapeutic regimes and identify pharmacokinetic parameters through non-linear regression analysis. It also explores using the properties of linear systems to identify non-linearities in the data, as well as the use of bootstrapping to compare parameter populations and handle multiple hypothesis tests. Finally, it shows how bootstrap values can estimate the distribution of derived parameters.

Revising Pharmacokinetics of Oral Drug Absorption: II Bioavailability-Bioequivalence Considerations

Pavlos Chryssafidis et al.

Summary: The study explores the application of PBFTPK models in assessing bioavailability and bioequivalence, developing a methodology for estimating absolute bioavailability exclusively from oral data. Simulated data showed that PBFTPK models are more similar to in vivo conditions, with useful parameters for assessing drug exposure and absorption extent. Estimates for F can be derived from oral data exclusively, with findings close to unity for theophylline formulations.
