Systemic administration of a pharmacologic iron chelator reduces cartilage lesion development in the Dunkin-Hartley model of primary osteoarthritis

Summary: The study demonstrates that deficiency of legumain attenuates acute renal tubular injury, inflammation, and ferroptosis in acute kidney injury models induced by ischemia-reperfusion injury or folic acid. Legumain deficiency prevents chaperone-mediated autophagy of GPX4, dampening ferroptosis induced by hypoxia or erastin. The results suggest that legumain promotes tubular ferroptosis in acute kidney injury by promoting chaperone-mediated autophagy of GPX4.

NCOA4-mediated ferritinophagy promotes ferroptosis induced by erastin, but not by RSL3 in HeLa cells

Summary: This study found that NCOA4 deficiency increases ferritin levels and makes cells more resistant to erastin but more sensitive to RSL3. Erastin promotes ferritinophagy, increasing free iron, lipid peroxidation, and sensitivity to ferroptosis, while RSL3 mechanism is independent of ferritin degradation process. The release of ferritin-iron in the execution of ferroptosis seems to depend on the inducing compound, its target, and downstream pathway of cell death activation.
