Coprophagy Prevention Decreases the Reproductive Performance and Granulosa Cell Apoptosis via Regulation of CTSB Gene in Rabbits

Summary: Coprophagy is an instinctive behavior in rabbits that has important effects on growth and reproductive performance. However, the underlying mechanism of this effect is still unknown. In this study, a coprophagy preventing model was used to evaluate the impact of preventing coprophagy on female rabbits. The results showed that preventing coprophagy did not affect feed intake but led to decreased body weight, organ weight, and complete reproductive failure during late pregnancy. RNA-seq analysis revealed that preventing coprophagy significantly affected the expression of 241 genes, with the majority being downregulated and involved in apoptosis, immune response, and metabolic pathways. The lysosomal cysteine protease cathepsin B (CTSB) was found to be significantly downregulated in the coprophagy prevention group and further experiments demonstrated that CTSB promotes the proliferation of rabbit granulosa cells and prevents apoptosis. Overexpression of CTSB increased secretion of progesterone and estradiol, while inhibition of CTSB decreased progesterone secretion. These findings suggest that preventing coprophagy has detrimental effects on reproduction, with CTSB playing a key role in the granulosa cells of the ovary.

Microbiome-Metabolome Analysis of the Immune Microenvironment of the Cecal Contents, Soft Feces, and Hard Feces of Hyplus Rabbits

Summary: The intestinal microbiota and their metabolites have important roles in rabbit growth and immune regulation. This study found significant differences in microbial community composition, cytokines, and SCFAs between soft feces and hard feces. Prevention of coprophagy led to changes in the cecal immune microenvironment, reducing the levels of anti-inflammatory cytokines and SCFAs, and affecting the expression levels of tight junction proteins associated with intestinal inflammation and barrier function.

These changes were attributed to decreased IGF-I and/or progesterone secretion, disruption of ovarian cell nuclei, and impaired ovarian cell response to FSH.
