A Formylglycine-Peptide for the Site-Directed Identification of Phosphotyrosine-Mimetic Fragments

Markus Tiemann et al.

Summary: Formylglycine is investigated as a molecular probe for the sensitive detection of fragments binding to a specific protein site. The formylglycine probe identified low-affinity fragments with high spatial resolution, and the best fragment hit was converted into a cellularly active, nanomolar inhibitor of the protein tyrosine phosphatase SHP2.

Catalyst-free site-selective cross-aldol bioconjugations

Nicholas D. J. Yates et al.

Summary: The bioconjugation of proteins to small molecules is crucial for studying biological mechanisms and developing biomaterials. This paper introduces a new method called spontaneous coupling via aldol ligation of proteins (SCALP), which is a catalyst-free, green, and site-selective protein ligation method. It demonstrates the utility of this method in targeting prostate cancer cells with functionalized nanobodies.

A Tale of Two Bioconjugations: pH Controlled Divergent Reactivity of Protein α-oxo-Aldehydes in Competing α-oxo-Mannich and Catalyst-Free Aldol Ligations

Tessa Keenan et al.

Summary: This study introduces two new methods for protein bioconjugation, utilizing small molecule aniline and/or phenol probes to modify alpha-oxo aldehyde handles within proteins. These methods offer control over the reaction selectivity under different pH conditions, resulting in bioconjugates with different functionalities.
