Selective Coupling of 1,2-Bis-Boronic Esters at the more Substituted Site through Visible-Light Activation of Electron Donor-Acceptor Complexes

Hui Wang et al.

Summary: In this study, a coupling reaction is reported that enables the reversal of traditional selectivity for functionalization of primary boronic esters. This is achieved through the formation of a boronate complex with an electron-rich aryllithium, followed by reaction with an electron-deficient aryl nitrile to form an electron donor-acceptor complex. Visible-light photoinduced electron transfer generates a primary radical, which isomerizes to a more stable secondary radical and undergoes radical-radical coupling with the arene radical-anion, resulting in the formation of beta-aryl primary boronic ester products. Furthermore, this method allows for stereodivergent coupling, providing trans-substituted products.

Photocatalyzed Decarboxylative Thiolation of Carboxylic Acids Enabled by Fluorinated Disulfide

Mikhail O. Zubkov et al.

Summary: This study describes the thiolation of carboxylic acids using a disulfide reagent with tetrafluoropyridinyl groups, which is mediated by light and an acridine-type photocatalyst. The method can be applied to various types of carboxylic acids and is useful for the late-stage modification of natural compounds and drugs. The formation of C-S bond is believed to be facilitated by the presence of the fluorinated pyridine fragment. The resulting sulfides can serve as redox-active radical precursors.

Heteroarylation of unactivated C-H bonds suitable for late-stage functionalization

Austin S. Miller et al.

Summary: The late-stage introduction of diverse heterocycles onto complex small molecules enables efficient access to new medicinally relevant compounds. Here, we report a system that enables direct C-H heteroarylation using a stable, commercially available O-alkenylhydroxamate with heterocyclic sulfone partners. The C-H heteroarylation proceeds efficiently with a range of aliphatic substrates and common heterocycles, and is a rare example of heteroarylation of strong C-H bonds. Importantly, the present approach is amenable to late-stage functionalization as the substrate is the limiting reagent in all cases.

Photoredox Fluoroalkylation of Hydrazones in Neutral and Reductive Modes

Boris A. van der Worp et al.

Summary: The visible light-promoted fluoroalkylation of hydrazones using 4-perfluoropyridine sulfides as fluoroalkyl radical sources was described. The process can proceed in neutral and reductive modes, delivering different products depending on the structure of starting substrates and reaction conditions. Ascorbic acid is used as a terminal reductant in the reductive process, recycling the photocatalyst and providing a source of hydrogen for nitrogen-centered radicals.

Using the Thiyl Radical for Aliphatic Hydrogen-Atom Transfer: Thiolation of Unactivated C-H Bonds

Liubov Panferova et al.

Summary: The metal- and catalyst-free thiyl-radical-mediated activation of alkanes involves thiolation of C-H bonds using tetrafluoropyridinyl disulfide under irradiation with 400 nm LEDs. The key C-H activation step is believed to proceed through hydrogen-atom abstraction by a fluorinated thiyl radical, allowing various types of C-H bonds to participate in the thiolation reaction. The resulting sulfides have great potential as photoredox-active radical precursors in reactions with alkenes and heteroarenes.

Visible-Light-Promoted Reversible Sulfide/Iodide Exchange in Fluoroalkyl Sulfides Enabled by Electron Donor-Acceptor Complex Formation

Boris A. van der Worp et al.

Summary: This study on the catalyst-free visible-light-mediated exchange of two photoredox active groups reveals a mechanism where appropriate choice of wavelength, solvent, and counterion allows preparative operations in both directions. The reactions involve electron donor-acceptor complexes and have been supported by UV/Vis absorption, NMR spectroscopies, and X-ray analysis.

Light-Mediated Sulfur-Boron Exchange

Liubov Panferova et al.

Summary: This study describes the synthesis of pinacol boronic esters through the reaction of sulfides bearing a tetrafluoropyridinyl group with bis(catecholato)diboron, followed by treatment with pinacol and triethylamine. The reaction is promoted by an organic photocatalyst (3DPA2FBN) under irradiation with 400 nm light, and is effective for primary, secondary, and tertiary sulfides. The electron depleting character of the fluorinated pyridine fragment is crucial in generating alkyl radicals.

Xanthates as Thiol Surrogates for Nucleophilic Substitution with Aryl Halides

Anatolii Sokolov et al.

Summary: The unprecedented xanthate-based protocol reported here allows for the preparation of aryl-alkyl thioethers in generally good yields and short reaction times by heating xanthates with aryl halides and cesium carbonate in methanol. This method enables the avoidance of contact with odorous thiols and the introduction of substituents that are virtually unavailable or inconvenient in use.

Decatungstate-Catalyzed C(sp3)-H Sulfinylation: Rapid Access to Diverse Organosulfur Functionality

Patrick J. Sarver et al.

Summary: The study presents a method for converting strong, aliphatic C(sp(3))-H bonds into alkyl sulfinic acids using decatungstate photocatalysis, which can be applied to a variety of C(sp(3))-rich molecules for efficient synthesis of complex sulfur-containing compounds. Furthermore, the method demonstrates potential for divergent synthesis of pharmaceutically relevant molecules through diversification of the sulfinic acid products into various medicinally relevant functional groups.

Radical Anion Promoted Chemoselective Cleavage of Csp2-S Bond Enables Formal Cross-Coupling of Aryl Methyl Sulfones with Alcohols

Jixiang Bai et al.

Summary: A novel formal cross-coupling of aryl methyl sulfones and alcohols to afford alkyl aryl ethers via an S(RN)1 pathway was developed, with the key step successfully utilized in the preparation of two marketed antitubercular drugs. The study revealed a dimsyl-anion initiated radical chain process as the major pathway, and DFT calculations indicated that the formation of a radical anion via nucleophilic addition of alkoxide to the aryl radical is crucial in determining the observed chemoselectivity.
