Selective covalent targeting of SARS-CoV-2 main protease by enantiopure chlorofluoroacetamide

Daiki Yamane et al.

Summary: This study reports an irreversible inhibitor of SARS-CoV-2 M-pro with chlorofluoroacetamide (CFA) as a covalent modification agent. The (R,R)-18 isomer showed higher inhibitory activity against M-pro than other isomers. The findings highlight the influence of CFA chirality on the covalent modification of proteinous cysteines and provide a basis for improving the potency and selectivity of CFA-based covalent inhibitors.

Iodine-Mediated C?C Double Bond Cleavage toward Pyrido[2,1-b]quinazolinones

Wei Hao et al.

Summary: This study describes a transition-metal-free C???C double bond cleavage reaction using molecular iodine as the catalyst. In the presence of K2CO3, the I2-mediated C???C bond cleavage followed by intramolecular annulation reaction can generate pyrido-[2,1-b]quinazolinones and related heterocyclic compounds with important biological activities.
