Physical and Functional Interaction of Mitochondrial Single-Stranded DNA-Binding Protein and the Catalytic Subunit of DNA Polymerase Gamma

Grzegorz L. Ciesielski et al.

Summary: This study reveals a direct interaction between mitochondrial single-stranded DNA-binding protein (mtSSB) and the catalytic subunit of mitochondrial replicative DNA polymerase, Pol gamma alpha, impacting DNA synthesis and exonucleolysis. The balance between attractive and repulsive affinities in the interaction between mtSSB and polymerase gamma holoenzyme (Pol gamma) plays a key role in these processes.

The mitochondrial single-stranded DNA binding protein is essential for initiation of mtDNA replication

Min Jiang et al.

Summary: mtSSB plays a crucial role in regulating mtDNA replication initiation by limiting transcription initiation to optimize RNA primer formation at both origins of mtDNA replication. This study also reveals that pathological versions of mtSSB in humans with severe mitochondrial disease are inefficient in supporting primer formation and initiation of mtDNA replication.

Multiprotein E. coli SSB ssDNA complex shows both stable binding and rapid dissociation due to interprotein interactions

M. Nabuan Naufer et al.

Summary: The study found that EcSSB binds to ssDNA in a biphasic manner, leading to incomplete wrapping at high concentrations. Destabilizing EcSSB wrapping through increased substrate tension, decreased substrate length, and protein mutation can disrupt the wrapping. When ssDNA is oversaturated, stimulated dissociation can rapidly remove excess EcSSB.
