Multiple causal variants underlie genetic associations in humans

Summary: This study evaluates the functional effects of genetic variants in high local linkage disequilibrium for independent cis-expression quantitative trait loci using a massively parallel reporter assay. The results show that 17.7% of the cis-expression quantitative trait loci exhibit multiple major allelic effects in tight linkage disequilibrium. The detected regulatory variants are highly enriched in activating chromatin structures and allelic transcription factor binding. Integration of the reporter assay profiles with colocalizations of cis-expression quantitative trait loci and complex traits across various human traits and diseases reveals sets of causal variants that contribute to genetic association signals.

Genome-wide functional screen of 3′UTR variants uncovers causal variants for human disease and evolution

Summary: The study developed a method called MPRAu, which was used to sensitively assay over 12,000 3'UTR variants. It revealed the regulatory activity of 3'UTRs, mainly attributed to simple sequences, and uncovered diverse molecular mechanisms at base pair resolution. Additionally, the MPRAu identified hundreds of 3'UTR causal variants and potential metabolic evolutionary adaptations in East Asians.
