Maf/ham1-like pyrophosphatases of non-canonical nucleotides are host-specific partners of viral RNA-dependent RNA polymerases

Adrian Valli et al.

Summary: Cassava brown streak disease (CBSD) is a serious threat to food security in Africa caused by two viruses, and this study reveals the role of ITPase in the infectivity of UCBSV in cassava. The study also demonstrates the partnership between RNA-dependent RNA polymerase (RdRP) and ITPase during infection. These findings contribute to understanding the interaction between the virus and the host.

Inosine triphosphate pyrophosphatase from Trypanosoma brucei cleanses cytosolic pools from deaminated nucleotides

Antonio E. Vidal et al.

Summary: This study presents the first report on a protozoan inosine triphosphate pyrophosphatase involved in the removal of harmful deaminated nucleotides from the cytosolic pool. TbITPA acts to exclude deaminated purines from the nucleoside triphosphate pools and influences the sensitivity of cells to IMP dehydrogenase inhibitors and antiviral drugs.

Disease-associated inosine misincorporation into RNA hinders translation

Jacob H. Schroader et al.

Summary: ITPase deficiency results in the accumulation of ITP, leading to misincorporation of inosine into RNA. This misincorporation perturbs translation, causing developmental anomalies and pathogenesis.

The isothiocyanate sulforaphane induces respiratory burst oxidase homologue D-dependent reactive oxygen species production and regulates expression of stress response genes

Andres Arruebarrena Di Palma et al.

Summary: Sulforaphane (SFN), a phytomolecule found in crucifers, exhibits anticancer properties in animals. This study investigated the signaling pathway of SFN in Arabidopsis and found that SFN induces the production of reactive oxygen species (ROS) through calcium-dependent activation of NADPH oxidase RBOH isoform D. RNA sequencing analysis revealed that SFN upregulates genes associated with heat shock proteins, oxidative stress response, and biotic and abiotic stress response in Arabidopsis.

The nucleotide metabolome of germinating Arabidopsis thaliana seeds reveals a central role for thymidine phosphorylation in chloroplast development

Markus Niehaus et al.

Summary: This study comprehensively analyzes the thymidylate metabolism during seed germination and seedling establishment in Arabidopsis. It reveals the important role of organelle thymidine salvage in thymidylate pool formation and the compensatory effect of cytosolic thymidylate synthesis in seedlings. The study also discovers a novel function of an organelle enzyme in removing noncanonical nucleotide triphosphates.

Viral inosine triphosphatase: A mysterious enzyme with typical activity, but an atypical function

Amy M. James et al.

Summary: In this study, it is found that plant-pathogenic viruses Cassava brown streak virus, Ugandan cassava brown streak virus, and Euphorbia ringspot virus encode a unique enzyme inosine triphosphatase not found in any other virus. However, the biological role of the encoded enzymes in virus infection is still unknown, showing potential for further research and understanding of cassava brown streak viruses.

Initiation of cytosolic plant purine nucleotide catabolism involves a monospecific xanthosine monophosphate phosphatase

Katharina J. Heinemann et al.

Summary: The study focuses on the dephosphorylation of XMP in plant cells and identifies an XMP phosphatase in Arabidopsis with specificity to XMP. The researchers demonstrate that XMPP initiates the catabolism of adenylate-derived nucleotides in vivo, while guanylates enter catabolism through a different pathway. The crystal structure and mutational analysis of XMPP provide insights into its high substrate specificity and efficient catalysis of the small XMP pool in vivo.

Arabidopsis thaliana endonuclease V is a ribonuclease specific for inosine-containing single-stranded RNA

Megumi Endo et al.

Summary: Endonuclease V is highly conserved across species and can cleave DNA and RNA containing inosine. AtEndoV in Arabidopsis thaliana exhibits variations in substrate specificity and prefers RNA over DNA substrates. This suggests that AtEndoV functions in processing RNA substrates with inosine induced by RNA damage, rather than A-to-I RNA editing.

Loss of MAR1 Function is a Marker for Co-Selection of CRISPR-Induced Mutations in Plants

Jannis Rinne et al.

Summary: The study describes the establishment of the knockout marker gene MAR1 for selection of CRISPR/Cas9-edited Arabidopsis seedlings and tomato explants in tissue culture. Mutations of MAR1 induced by the CRISPR system confer kanamycin-resistance to Arabidopsis plants and tomato tissues. Co-selection of a mutation in a second target gene at higher frequency has been observed for the first time in plants, indicating the potential utility of this technique for selecting edited plants.

Enhanced nucleotide analysis enables the quantification of deoxynucleotides in plants and algae revealing connections between nucleoside and deoxynucleoside metabolism

Henryk Straube et al.

Summary: A new method was developed to detect and quantify low-abundance (deoxy)ribonucleotides and (deoxy)ribonucleosides in plants with high sensitivity and robustness. The study revealed the intricate interconnection between deoxyribonucleotide metabolism and ribonucleoside catabolism, as well as the correlation between nucleotide ratios and genomic contents across different plant species. The findings demonstrate the potential of this method in providing valuable insights into plant nucleotide metabolism.
