Methyl beta-D-galactopyranoside esters as potential inhibitors for SARS-CoV-2 protease enzyme: synthesis, antimicrobial, PASS, molecular docking, molecular dynamics simulations and quantum computations

Md R. Amin et al.

Summary: Carbohydrate esters play an important role in medicinal chemistry for the synthesis of biologically active drugs. In this study, methyl beta-D-galactopyranoside (MGP) was reacted with various acyl halides to produce 6-O-acyl MGP esters, which were further modified into 2,3,4-tri-O-acyl MGP esters. The newly synthesized compounds showed promising antifungal functionality compared to their antibacterial activities. Among them, compounds 3 and 10 exhibited the strongest inhibitory effects against Bacillus subtilis and Escherichia coli. Density functional theory calculations and molecular docking revealed the properties and interactions of these compounds. Molecular dynamics simulation validated the binding affinities obtained from molecular docking. The pharmacokinetic characterization demonstrated that these MGP esters have improved kinetic properties. Structure activity relationships study indicated that the acyl chain and chemical groups influenced the activity of the esters. Overall, this comprehensive computational and statistical analysis suggests that these selected MGP esters could be potential inhibitors against SARS-CoV-2.

Linking azoles to isoniazid via hydrazone bridge: Synthesis, crystal structure determination, antitubercular evaluation and computational studies

Ebru Kocak Aslan et al.

Summary: The emergence of drug-resistant strains of Mycobacterium tuberculosis (Mtb) has complicated efforts to eliminate tuberculosis. Developing new antitubercular drugs is crucial in addressing this challenge. Researchers used molecular hybridization to link the frontline drug isoniazid (INH) with various azole rings. These compounds showed excellent antimycobacterial activity and low cytotoxicity.

Recent advances in the medicinal chemistry of carbonic anhydrase inhibitors

Shubham Kumar et al.

Summary: Carbonic anhydrase is an important enzyme involved in various physiological and pathological processes, with 16 different isoforms in humans. Inhibitors targeting different isoforms of carbonic anhydrase have clinical applications in treating various diseases, but current drugs lack selectivity leading to undesired side effects. Efforts are being made to develop novel isoform-selective inhibitors of carbonic anhydrase for therapeutic implications.

Design, synthesis and biochemical evaluation of novel carbonic anhydrase inhibitors triggered by structural knowledge on hCA VII

Francesca Mancuso et al.

Summary: To address the challenge of isoform selectivity, the entrance of the cavity for certain druggable human Carbonic Anhydrases (hCAs) was explored. Using X-ray crystallographic studies, a series of 4-(4(hetero)aroylpiperazine-1-carbonyl)benzene-1-sulfonamides was developed based on complex with hCA VII. Through docking simulations, the capability of newer benzenesulfonamides to fit the hCA VII catalytic cavity was evaluated. Subsequently, a series of thirteen benzenesulfonamides was synthesized and tested, with the 4-(4-(furan-2-carbonyl)piperazine-1-carbonyl)benzenesulfonamide showing significant affinity towards hCA VII and good selectivity over hCA I when compared to Topiramate (TPM).

Design, synthesis, antibacterial activity evaluation and molecular modeling studies of new sulfonamides containing a sulfathiazole moiety

Tugba Meseli et al.

Summary: Sulfonamides, as the oldest synthetic antibacterial agents, have been compromised due to widespread resistance. Research on modified sulfathiazole derivatives has shown new compounds with better antibacterial activities against Staphylococcus aureus, particularly when introducing bulky lipophilic substituents. However, the mechanisms behind these antibacterial activities are still not completely understood.
