Broad white matter impairment in multiple system atrophy

Summary: Multiple system atrophy (MSA) is a rare neurodegenerative disorder characterized by widespread accumulation of alpha-synuclein, primarily in oligodendrocytes. Whole brain deep and superficial white matter diffusivity abnormalities were observed in MSA patients but not in Parkinson's disease (PD) patients. These abnormalities were associated with motor and cognitive functions in MSA patients.

Characteristics of Epigenetic Clocks Across Blood and Brain Tissue in Older Women and Men

A Meta-Analysis of Brain DNA Methylation Across Sex, Age, and Alzheimer's Disease Points for Accelerated Epigenetic Aging in Neurodegeneration

Summary: AD is characterized by specific brain DNA methylation patterns, affected by age and sex. This study analyzed DNAm datasets from healthy adults and AD patients to identify sex-, age-, and AD-associated epigenetic profiles. Results showed shared DNAm differences between males and females across brain regions, with aging affecting cortical regions differently from the cerebellum. AD-related epigenetic modifications were enriched in age-related probes, suggesting accelerated epigenetic aging in the disease.

The association of epigenetic clocks in brain tissue with brain pathologies and common aging phenotypes

Summary: Epigenetic clocks, calculated by combining DNA methylation states across select CpG sites to estimate biologic age, were used in brain tissue of older participants to explore aging phenotypes. The Cortical clock showed stronger associations with Alzheimer's disease pathology compared to other established clocks, and was also significantly related to clinical phenotypes such as dementia and cognitive decline.

White matter alterations in Parkinson's disease with levodopa-induced dyskinesia

Summary: This study aimed to investigate white matter alterations in Parkinson's disease patients with and without levodopa-induced dyskinesia using advanced diffusion magnetic resonance imaging techniques. The results showed significant differences in white matter microstructural disruption, especially in temporal lobe fibers, between patients with and without levodopa-induced dyskinesia. The tracts in temporal lobe fibers had a more than 2-fold odds ratio for the presence of levodopa-induced dyskinesia, suggesting a potential association with the pathogenesis of the sequela.
