Animal models of brain-first and body-first Parkinson's disease

Nathalie Van Den Berge et al.

Summary: Alpha-synuclein aggregates are the hallmark of Parkinson's disease and can propagate along the brain-body axis in a stereotypical pattern. The heterogeneity of symptoms and pathology in Parkinson's disease patients may be explained by the variable onset site, either in the brain or body. Animal models have been developed to reproduce Parkinson's disease pathology in order to study the site of disease onset.

Alpha-Synuclein-Specific Naturally Occurring Antibodies Inhibit Aggregation In Vitro and In Vivo

Anne K. Braczynski et al.

Summary: Parkinson's disease is associated with changes in the abundance of naturally occurring antibodies (nAbs) against alpha-synuclein (α Syn). In this study, nAbs-alpha Syn were found to specifically bind monomeric α Syn and inhibit its aggregation in vitro. Additionally, the addition of nAbs-alpha Syn to cultured cells affected intracellular α Syn aggregates.

ABBV-0805, a novel antibody selective for soluble aggregated α-synuclein, prolongs lifespan and prevents buildup of α-synuclein pathology in mouse models of Parkinson's disease

Eva Nordstrom et al.

Summary: ABBV-0805 is a humanized monoclonal antibody with high selectivity for aggregated alpha-synuclein, demonstrating efficacy in cellular and animal models, and showing potential disease-modifying treatment for Parkinson's disease based on strong preclinical findings.

Administration of AAV-Alpha Synuclein NAC Antibody Improves Locomotor Behavior in Rats Overexpressing Alpha Synuclein

Yun-Hsiang Chen et al.

Summary: The study demonstrates that intervention with AAV-NAC32 on alpha-Synuclein in animals can improve PD-like symptoms, including reducing bradykinesia and restoring tyrosine hydroxylase loss. This research provides hope for this intervention technique as a potential therapeutic candidate for PD.
