The amyloid hypothesis in Alzheimer disease: new insights from new therapeutics

Early-onset and late-onset AD exhibit different genetic characteristics, with late-onset showing high genetic heterogeneity. Presenilin 2 N141I mutation induces hyperactive immune response through the epigenetic repression of REV-ERBα

Summary: This study demonstrates that the Presenilin 2 N141I mutation causes the overproduction of clock-controlled cytokines and memory deficits through hypermethylation-mediated suppression of the REV-ERB alpha gene. The mutation leads to excessive immune cell activation and cognitive decline in response to mild immune challenges. Treatment with the antipsychotic chlorpromazine restores the REV-ERB alpha level and prevents cognitive decline in mice. These findings reveal a pathogenic link between this AD mutation and immune cell overactivation through epigenetic suppression of REV-ERB alpha.

Rare missense mutations in ABCA7 might increase Alzheimer's disease risk by plasma membrane exclusion

Summary: This study investigated the contribution and pathogenicity of rare missense, indel and splice variants in the ABCA7 gene in Belgian Alzheimer's disease (AD) patient and control cohorts. The results suggest that rare ABCA7 missense mutations can contribute to AD by inducing protein mislocalization, resulting in a lack of functional protein at the plasma membrane. Additionally, some of the observed missense mutations affect amino acid residues that are conserved in ABCA1 and ABCA4 and correspond to disease-causing mutations in Tangier and Stargardt disease.
