Rapid Regulation of Glutamate Transport: Where Do We Go from Here?

Alain M. Guillem et al.

Summary: Glutamate, the predominant excitatory neurotransmitter in the mammalian central nervous system, is regulated by signals and post-translational modifications through a family of transporters. Abnormal glutamatergic signaling has been linked to various psychiatric and neurological disorders, but understanding the mechanisms involved in regulation will be crucial for further advancements in the field.

Enhanced Feedback Inhibition Due to Increased Recruitment of Somatostatin-Expressing Interneurons and Enhanced Cortical Recurrent Excitation in a Genetic Mouse Model of Migraine

Ivan Marchionni et al.

Summary: Migraine is a complex brain disorder characterized by unilateral headache and dysfunction in multisensory information processing. This study investigates the cellular and circuit mechanisms underlying familial hemiplegic migraine (FHM) in mouse models. The findings suggest that enhanced excitatory neurotransmission and unaltered inhibitory neurotransmission may contribute to the pathogenesis of FHM.

Optogenetic Spreading Depression Elicits Trigeminal Pain and Anxiety Behavior

Andrea M. Harriott et al.

Summary: The study used a minimally invasive optogenetic method to induce SD and found that a single SD caused bilateral periorbital mechanical allodynia, while repeated SDs also produced bilateral periorbital mechanical allodynia that lasted for 4 days. Repeated SDs increased anxiety behavior and facial expression pain scores, but did not affect working memory.

Non-canonical glutamate signaling in a genetic model of migraine with aura

Patrick D. Parker et al.

Summary: A study on mice with familial hemiplegic migraine type 2 (FHM2) mutation found slower clearance during sensory processing and spontaneous plumes of glutamate. These plumes, predicted by elevated basal glutamate levels and plume frequency, provide a novel mechanism for migraine with aura onset.

Regulation of Glutamate, GABA and Dopamine Transporter Uptake, Surface Mobility and Expression

Renae M. Ryan et al.

Summary: Neurotransmitter transporters play a crucial role in regulating neurotransmitter concentration and can be dynamically regulated through redistribution and stabilization. This allows for adjustments in uptake capacity for different neurotransmitters in response to changes in neuronal activity, blood flow, and cell-to-cell interactions.

Specific activation of GluN1-N2B NMDA receptors underlies facilitation of cortical spreading depression in a genetic mouse model of migraine with reduced astrocytic glutamate clearance

Giovanna Crivellaro et al.

Summary: Migraine is a common but poorly understood sensory circuit disorder. Mouse models of familial hemiplegic migraine show increased susceptibility to cortical spreading depression, which is mainly caused by reduced expression of astrocytic Na+, K+-ATPases. The enhanced NMDAR activation in FHM2 mice is activity-dependent and can be rescued by inhibition of GluN1-N2B NMDARs.
