Rewired glycosylation activity promotes scarless regeneration and functional recovery in spiny mice after complete spinal cord transection

Summary: The spiny mouse is capable of spontaneous and fast restoration of function after severe spinal cord injury (SCI) due to the formation of scarless regenerative tissue. Transcriptomic analysis revealed the rewiring of glycosylation biosynthetic pathways in the spiny mouse, leading to pro-regenerative proteoglycan signature at the SCI site. This study highlights the importance of glycosylation switch in axon regeneration after SCI.

Crosstalk between epitranscriptomic and epigenetic mechanisms in gene regulation

Summary: Epigenetic modifications play a crucial role in gene expression regulation through modifications on genomic DNA, histones, and more recently, mRNA. These modifications, especially the common N6-methyladenosine (m6A) methylation, not only influence RNA stability and translation efficiency, but also interact with histone/DNA epigenetic mechanisms to specify transcriptional outputs and recruit chromatin modifiers. This review focuses on dissecting the feedback circuits orchestrated by m6A, which regulate histone modifications and the activity of regulatory RNAs, such as long noncoding RNA and chromosome-associated regulatory RNA. The evidence suggests that m6A acts as a versatile checkpoint linking different layers of gene regulation.

The brain mitochondrial kinase P21-activated kinase 5 (PAK5) is activated in axons in response to ischemic stress and axonal injury, leading to the remobilization and replacement of damaged mitochondria. This mechanism protects axon survival and facilitates regeneration in both in vitro and in vivo models, supporting central nervous system (CNS) survival and regeneration.
