Summary: A convenient method for preparing high-quality microcrystals of a bacterial quinoprotein enzyme has been reported, enabling high-resolution X-ray crystallography studies. This method involves mechanically crushing large crystals, seeding particles, and allowing microcrystals to grow naturally, leading to efficient microcrystal formation. The damage-free structure determined at 2.2 angstrom resolution through serial femtosecond X-ray crystallography was essentially identical to that determined by cryogenic crystallography using synchrotron X-ray radiation.
