Paradoxical somatodendritic decoupling supports cortical plasticity during REM sleep

Summary: During REM sleep, somatodendritic decoupling in pyramidal neurons of the prefrontal cortex occurs, mainly driven by neurons from the central medial thalamus. This decoupling promotes opposite synaptic plasticity mechanisms that optimize emotional responses to future behavioral stressors.

Is REM sleep a paradoxical state?: Different neurons are activated in the cingulate cortices and the claustrum during wakefulness and paradoxical sleep hypersomnia

Summary: Using TRAP2 transgenic mice, different neurons were found to be activated during PS and W in the anterior cingulate and rostral and caudal retrosplenial cortices as well as the claustrum. Additionally, during PS, the distribution of neurons in the rRSP and cRSP was limited to superficial layers, in contrast to the widespread activation across all layers during W. These results clearly demonstrate at the cellular level that PS and W are two completely different states in terms of neocortical activation.

Summary: A unique neuron type called down state-active (DSA) neurons has been identified in mice and rats, which show anti-correlated spiking activity with principal cells and interneurons during non-REM sleep, especially during the down state. Despite their sparsity, DSA neurons play critical roles in physiological functions, shaping the firing sequence of neurons during down-up transitions and interfering with memory consolidation during NREM sleep.

Theta oscillations and sharp-wave/ripples tightly controlled the firing of projection neurons in a target region-specific manner.
