Overview of current adipose-derived stem cell (ADSCs) processing involved in therapeutic advancements: flow chart and regulation updates before and after COVID-19

Loubna Mazini et al.

Summary: Adipose-derived stem cells have low immunogenicity and various self-renewal abilities, can differentiate into different tissue-specific progenitors, migrate into damaged sites, and act through autocrine and paracrine pathways. However, the lack of standardization protocols and discrepancies in cell outcomes and engraftment pose limitations on their widespread use.

Safe harbor-targeted CRISPR-Cas9 homology-independent targeted integration for multimodality reporter gene-based cell tracking

John J. Kelly et al.

Summary: The study utilized HITI CRISPR-Cas9 minicircle donors for precise targeted knock-in of reporter genes at safe harbor loci. Results showed higher knock-in efficiency and functional reporter gene activity with HITI vectors, enabling multi-modal longitudinal in vivo imaging of cells. This work demonstrates the first CRISPR-Cas9 HITI system for large DNA donor constructs at a safe harbor locus.

Human Red Blood Cells Modulate Cytokine Expression in Monocytes/Macrophages Under Anoxic Conditions

Antonella Antonelli et al.

Summary: This study investigated the effect of oxygen tension on stem and progenitor cell proliferation, differentiation, and cytokine expression in the bone marrow hematopoietic niche. Results showed that the presence of red blood cells up-regulated IL-8 mRNA while IL-6 and VEGF mRNA expression appeared down-regulated, and administration of RBCs led to an increase in TNF alpha expression levels in MonoMac 6 cells, suggesting a potential hypoxia-independent modulation of these factors. Further studies should explore these preliminary observations in more detail considering the role of oxygen in the hematopoietic niche.

Short lifespan of syngeneic transplanted MSC is a consequence of in vivo apoptosis and immune cell recruitment in mice

Mihai Bogdan Preda et al.

Summary: The study found that transplanted MSC had limited migration capacity and were short-lived, disappearing almost completely within a week of transplantation regardless of the administration route. The transplanted MSC activated hypoxia signaling pathways, followed by apoptosis and subsequent engulfment by macrophages.

[18F]FDG-labelled stem cell PET imaging in different route of administrations and multiple animal species

Naoko Nose et al.

Summary: Stem cell therapy shows promising potential for tissue regeneration and cancer treatment, while non-invasive cell tracking can offer important insights into in vivo cell distribution. This study demonstrated the feasibility of using PET imaging and in vitro cell labelling with [18F]FDG to monitor engrafted stem cells in multiple animal models, providing valuable information for understanding and optimizing stem cell therapy.
