Summary: This study demonstrates that co-inhibiting hyperactivated XBP1 and redundant intracellular ROS through KT-NE can effectively reduce lipid accumulation in dysfunctional lipid-laden TIDCs, leading to the reactivation of TIDCs and restoration of their capacity to expand and stimulate tumor-specific T cells. Additionally, both KT-NE-treated TIDCs (adoptive transfer) and KT-NE itself can significantly improve the efficacy of DC-mediated immunotherapy and reduce tumor burden in malignant ovarian cancer (OvCa)-bearing mice.

Effects of tocilizumab and dexamethasone on the downregulation of proinflammatory cytokines and upregulation of antioxidants in the lungs in oleic acid-induced ARDS

Summary: This study aimed to investigate the effects of tocilizumab and dexamethasone on oxidative stress, antioxidant parameters, and cytokine storms in acute lung injury caused by oleic acid. The results showed that tocilizumab and dexamethasone effectively prevented cytokine storms by downregulating the expression of proinflammatory cytokines. They also upregulated antioxidant parameters and exhibited protective effects against cell damage caused by oleic acid.

Summary: Collagen XV is highly expressed in adipose tissue and is related to extracellular matrix remodeling, fibrosis, and inflammation in adipose tissue. Overexpression of Col XV in mice leads to infiltration of macrophages into white adipose tissue, exacerbating inflammation and ER stress. Inhibition of the Integrin beta 1/FAK pathway can reverse the effects of Col XV on ER stress and inflammation in adipocytes.

Targeted regulation of lymphocytic ER stress response with an overall immunosuppression to alleviate allograft rejection

Summary: Regulating lymphocytic ER unfolded protein response (UPR) could be a potential therapeutic breakthrough in alleviating graft rejection. These findings suggest that lymphocytic IRE1 alpha inactivation attenuates transplant rejection and prolongs graft survival, with limited effector function and memory commitment of host T-8. The combination of IRE1 alpha inhibition with immunosuppressant tacrolimus (FK506) shows an even higher immunosuppressive effect through synergistic regulation of inflammatory transcription factors.
