Design, Synthesis and Antiproliferative Evaluation of Bis-Indole Derivatives with a Phenyl Linker: Focus on Autophagy

Design, synthesis and pharmacological evaluation of β-carboline derivatives as potential antitumor agent via targeting autophagy

Summary: A series of novel beta-carboline derivatives were designed, synthesized and evaluated for their potential as anticancer agents. Compound 6g exhibited the most potent antiproliferative activity against three different cancer cell lines, with IC50 values ranging from 2.02 to 3.86 μM. In vivo studies showed that compound 6g suppressed tumor development and reduced tumor weight in a mouse colorectal cancer model. Further investigations revealed that compound 6g inhibited HCT116 cell growth by activating the ATG5/ATG7-dependent autophagic pathway. These molecules have potential as candidates for the development of colorectal cancer therapy agents.

Summary: The newly synthesized indole chalcone derivative ZK-CH-11d has been found to have an inhibitory effect on breast cancer cells and minimal effect on non-cancer cells. It can suppress cell proliferation by causing cell cycle arrest and inducing apoptosis. In addition, ZK-CH-11d can activate autophagy, but it is not the principal factor contributing to the anti-proliferative effect.

The Anticancer Potential of Doxycycline and Minocycline-A Comparative Study on Amelanotic Melanoma Cell Lines

Summary: This study found that minocycline and doxycycline have anti-melanoma effects on amelanotic melanoma cells, including inhibition of cell proliferation, decreased cell viability, induction of apoptosis, and changes in cell cycle profile. The study also found that these drugs increase the level of LC3A/B, an autophagy marker. Overall, doxycycline was found to be more potent than minocycline.

Summary: The global cancer burden in 2020 saw an estimated 19.3 million new cancer cases and almost 10.0 million cancer deaths. Female breast cancer surpassed lung cancer as the most commonly diagnosed cancer, while lung cancer remained the leading cause of cancer death. These trends are expected to rise in 2040, with transitioning countries experiencing a larger increase compared to transitioned countries due to demographic changes and risk factors associated with globalization and a growing economy. Efforts to improve cancer prevention measures and provision of cancer care in transitioning countries will be crucial for global cancer control.
