Polyhydroxyalkanoates from a Mixed Microbial Culture: Extraction Optimization and Polymer Characterization

Summary: Polyhydroxyalkanoates (PHA) are potential substitutes for conventional oil-based plastics, but high production costs are a limiting factor. This study investigated the extraction of poly(3-hydroxybutyrate-co-3-hydroxyvalerate) using sodium hydroxide or sodium hypochlorite as digestion agents. The optimized conditions resulted in high purity and recovery rates for PHA.

Extraction of low molecular weight polyhydroxyalkanoates from mixed microbial cultures using bio-based solvents

Summary: In this study, the molecular weight of PHBV was intentionally reduced to facilitate extraction and handling, and the use of bio-based solvents 2-MTHF and cyrene for the extraction of low MW PHBV was investigated. The results showed that these new solvents had higher extraction efficiency for low MW PHBV compared to benchmark solvents chloroform and DMC.

Biosynthesis and Properties of a P(3HB-co-3HV-co-4HV) Produced by Cupriavidus necator B-10646

Summary: The synthesis of P(3HB-co-3HV-co-4HV) copolymers by Cupriavidus necator B-10646 using fructose or sodium butyrate as the main C-substrate with the addition of gamma-valerolactone as a precursor of 3HV and 4HV monomers was studied. The physicochemical properties of the copolymers and films were analyzed using various methods, showing changes depending on the proportions of monomers.

Subcritical Water as a Pre-Treatment of Mixed Microbial Biomass for the Extraction of Polyhydroxyalkanoates

Downstream processing and characterization of polyhydroxyalkanoates (PHAs) produced by mixed microbial culture (MMC) and organic urban waste as substrate

Summary: The research investigated the utilization of food waste and sewage sludge as organic substrate for the synthesis of microbial PHAs at pilot scale. Two stabilization methods, thermal drying and wet acidification, were found for preserving the amount of intracellular PHA produced by open mixed microbial culture. The extracted PHA from wet-acidified biomass had higher average molecular weights compared to PHA extracted from thermally stabilized dried biomass.

Results showed that larger screw speeds promote clay particle break-up, while smaller screw speeds favor tactoid erosion. Lowering the PDMS viscosity in the first mixing zone of the screw facilitates dispersion at lower screw speeds.

Ethylic Esters as Green Solvents for the Extraction of Intracellular Polyhydroxyalkanoates Produced by Mixed Microbial Culture

Summary: This study focused on the extraction of poly(3-hydroxybutyrate-co-3-hydroxyvalerate) produced by mixed microbial culture using ethyl esters. Among the tested ethyl esters, ethyl acetate was found to be the best solvent for dissolving the copolymer at the lowest temperature. The study showed that extraction at different temperatures can affect the recovery yield, polymer purity, and molecular weight of the extracted polyhydroxyalkanoates.

Combined Strategies to Boost Polyhydroxyalkanoate Production from Fruit Waste in a Three-Stage Pilot Plant
