Novel piperine-carboximidamide hybrids: design, synthesis, and antiproliferative activity via a multi-targeted inhibitory pathway

Novel indazole derivatives as potent apoptotic antiproliferative agents by multi-targeted mechanism: Synthesis and biological evaluation

Summary: Indazole is an important class of heterocyclic compounds with diverse biological activity. In this study, a novel series of differently substituted indazole derivatives 6a-v were synthesized and evaluated for their biological activity. The results demonstrated that compounds 6f, 6i, 6j, 6s, and 6n exhibited significant antiproliferative activity against cancer cells and showed promising potential as potent antiproliferative agents by inhibiting EGFR, CDK2, and c-Met and inducing apoptosis through cytochrome C overexpression and activation of the intrinsic apoptotic pathway.

Summary: A new series of quinazoline-4-one/chalcone hybrids, 7-26, were synthesized and tested as EGFR inhibitors with antiproliferative activity. Three compounds showed the greatest antiproliferative activity and were the most potent EGFR inhibitors. These three compounds improved the levels of active caspase-3, 8, and 9, induced cytochrome c and Bax levels, and down regulated Bcl_2 levels. The most active inhibitors docked well inside EGFR active sites.

Compounds 5d, 5e, and 5f exhibited the strongest antiproliferative activity and showed potent inhibition against CDK2 and EGFR. The compounds were also found to increase the activity of caspases and cytochrome C levels. Molecular docking studies confirmed the strong binding affinity of compound 5h to the active sites of CDK2 and EGFR.

The compounds exhibited high binding affinities toward tubulin, suggesting it as a possible target for these compounds.

Summary: A novel series of tri-aryl imidazole derivatives carrying benzene sulfonamide moiety were designed for their selective inhibitory activity against hCA IX and XII. Among them, six compounds showed potent and selective CA IX inhibition, with 5g and 5b demonstrating higher antiproliferative activity compared to other tested compounds. Docking studies of these two compounds revealed their favorable binding interactions with CA-IX, similar to that of ligand 9FK. Molecular modeling simulations supported the biological evaluation results.
