A stepwise strategy integrating metabolomics and pseudotargeted spectrum-effect relationship to elucidate the potential hepatotoxic components in Polygonum multiflorum

Yunfei Song et al.

Summary: This study developed a strategy combining metabolomics and pseudotargeted spectrum-effect relationship to explore the potential hepatotoxic components in Polygonum multiflorum. By identifying and comparing 112 components, as well as screening the reduced differential components between raw and processed PM, 16 components with potential hepatotoxicity were identified. Mathematical models were also established to further identify the key variables affecting hepatotoxicity.

Summary: This study aimed to identify potential biomarkers of hepatotoxicity induced by Polygonum Multiflorum Radix Preparata (PMP) using metabolomics analysis. The results revealed that PMP-induced liver injury may be related to cholestasis and dysregulated lipid metabolism. The metabonomic method could be a valuable tool for clinical diagnosis of PMP-induced liver injury.

Summary: Phellodendri Chinese Cortex has been traditionally used for treating hyperuricemia and gout. Berberine (BBR), its active component, has been proven effective in reducing gout inflammation. Dihydroberberine (DHB), a derivative of BBR, showed significant hypouricemic and renoprotective effects in hyperuricemic mice, possibly by inhibiting XOD, URAT1, GLUT9, and NLRP3 inflammasome activation in the kidney.

CD73 Maintains Hepatocyte Metabolic Integrity and Mouse Liver Homeostasis in a Sex-Dependent Manner

Karel P. Alcedo et al.

Summary: The study demonstrates that hepatocyte CD73 supports long-term metabolic liver homeostasis through regulating the AMPK signaling pathway in a sex-dependent manner. CD73-LKO mice develop liver disease after 21 weeks, with more severe symptoms observed in male mice. These findings have implications for human liver diseases marked by CD73 dysregulation.

Investigation of the idiosyncratic hepatotoxicity of Polygonum multiflorum Thunb. through metabolomics using GC-MS

Yan Lin et al.

Summary: The study identified that Polygonum multiflorum ethanol extract, 50% ethanol elution fractions, and tetrahydroxystilbene glucoside from PM can induce idiosyncratic hepatotoxicity, with TSG being the main hepatotoxic component. Changes in 40 endogenous metabolites in the rat liver and 32 types of endogenous metabolites in rat serum were identified. Different biomarkers were linked to disruptions in amino acid, fatty acid, oxalate, energy, glucose, and fat metabolism induced by PM and TSG. The mechanism of idiosyncratic hepatotoxicity in PM involves disruptions in amino acid metabolism, lipid metabolism, energy metabolism, and glucose metabolism induced by TSG.
